The disinfection method of seed and culture condition of aseptic seedling in tissue culture of Tetracentron sinensis were optimized in this experiment.There were 4 disinfection methods used in total and the results were as follows:the germination of T. sinensis seed was inhabited strongly by 0.1% HgCl₂ and weakly by 4% NaClO;the best disinfection method was 4% NaClO.The culture medium with moderate salt and low nitrogen was in favor of the germination of seed,and 1/4 MS medium was the optimal medium for culturing aseptic seedling of T. sinensis.
