Molecular Identification of Polygonatum cyrtonema and P. sibiricum in Hubei Region Based on ETS Sequences

基于ETS序列对湖北地区多花黄精与黄精的分子鉴定

Abstract: The external transcribed spacer (ETS) molecular marker was used to identify and analyze Polygonatum cyrtonema and P. sibiricum from Hubei Province, to provide a basis for their efficient discrimination. A total of five individuals of P. cyrtonema and eleven of P. sibiricum were collected from Hubei Province for molecular analysis. The ETS sequence was amplified by PCR, followed by sequence feature analysis using bioinformatics methods. A phylogenetic tree was constructed using the neighbor-joining (NJ) method. Results showed that the ETS sequences of both species were 1 000 bp in length, yet their nucleotide compositions differed markedly. In P. cyrtonema, GC content was 41.6%-45.1%, with an average intraspecific genetic distance of 0.208 and an intraspecific sequence similarity of 88.1%, containing 134 parsimony informative sites. In P. sibiricum, GC content was significantly higher by 56.5%-61.5%, the average intraspecific genetic distance was 0.131, and the sequence similarity was 79.2%, with 298 parsimony informative sites. In the NJ phylogenetic tree, P. cyrtonema and P. sibiricum formed clearly separated clades, enabling effective discrimination. The ETS sequence can serve as a species-specific molecular marker for distinguishing P. cyrtonema and P. sibiricum, providing a molecular basis for precise germplasm identification and genetic diversity analysis of the two species, as well as for quality control of Hubei's local Polygonatum medicinal materials.

摘要: 利用外转录间隔区(ETS)分子标记,对湖北地区多花黄精Polygonatum cyrtonema与黄精P. sibiricum进行鉴定分析,为两种植物的高效鉴别提供依据。以湖北地区5份多花黄精和11份黄精为研究材料,通过PCR扩增获得ETS序列,结合生物信息学方法进行序列特征分析,并利用邻接法(Neighbor-joining,NJ)构建系统发育树。结果表明,两种黄精的ETS序列长度平约1000 bp,但核苷酸组成差异显著——多花黄精GC含量为41.6%~45.1%,种内平均遗传距离为0.208,种内序列相似度为88.1%,含134个简约信息位点;黄精GC含量显著升高(56.5%~61.5%),种内平均遗传距离为0.131,序列相似度79.2%,简约信息位点298个。邻接系统发育树中,多花黄精与黄精形成明显独立分支,实现有效区分。ETS序列可作为多花黄精与黄精种间鉴定的特异性分子标记,为二者的种质资源精准鉴定、遗传多样性分析及湖北道地黄精药材的质量控制提供分子依据。
