金山芋淀粉对小鼠急性胃黏膜损伤的保护作用

Protective Effect of Starch from Canna indica 'Edulis' against Acute Gastric Mucosal Injury in Mice

摘要: 为探究产自广东江门的金山芋Canna indica 'Edulis' 淀粉对小鼠急性胃黏膜损伤的保护作用,本研究构建了盐酸-乙醇小鼠急性胃黏膜损伤模型,通过灌胃金山芋淀粉,对小鼠的胃指数、胃组织病理学、胃黏膜损伤、血清中脂质代谢以及胃组织中氧化应激水平变化进行观察与测定。结果显示,与模型组对比,金山芋淀粉给药组小鼠胃指数显著升高,胃黏膜损伤指数显著降低,其中金山芋淀粉高剂量组胃黏膜损伤积分指数降低到2.70,损伤抑制率高达60.51%。胃组织病理学结果显示,金山芋给药组小鼠胃表面颜色和形态逐渐趋于正常组小鼠,与模型组对比,出血带的长度和宽度均显著减少,同时胃黏膜充血、出血性损伤以及炎性细胞浸润明显改善,损伤面积明显减少,上皮细胞结构较为完整。此外,与模型组对比,金山芋给药组小鼠血清中胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量明显降低,高密度脂蛋白胆固醇(HDL-C)含量明显升高,胃组织中总超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)的活性明显提升。由此可见,金山芋淀粉通过改善盐酸-乙醇诱导的小鼠胃组织氧化应激状态,调节脂质代谢紊乱,协同减轻胃黏膜充血、出血及炎性损伤,从而发挥对小鼠急性胃黏膜损伤的保护作用。本研究为开发金山芋淀粉保护胃黏膜损伤产品提供了科学依据。

Abstract: To investigate the protective effect of starch from Canna indica 'Edulis' (Jinshanyu) produced in Jiangmen, Guangdong Province, a mouse model of acute gastric mucosal injury was induced by hydrochloric acid-ethanol. By intragastric administration of Jinshanyu starch, the gastric index, gastric histopathology, gastric mucosal injury, serum lipid metabolism, and changes in oxidative stress levels in gastric tissue of mice were observed and determined. The results showed that, compared with the model group, the gastric index of mice in the Jinshanyu starch treatment groups were significantly increased, while the gastric mucosal injury index was significantly decreased. Specifically, the gastric mucosal injury score index of the high-dose Jinshanyu starch group was reduced to 2.70, with an injury inhibition rate as high as 60.51%. Histopathological results of gastric tissue indicated that the color and morphology of the gastric surface of mice in the Jinshanyu treatment groups gradually approached those of the normal group. In comparison with the model group, the length and width of the hemorrhagic bands were significantly reduced; meanwhile, gastric mucosal congestion, hemorrhagic injury, and inflammatory cell infiltration were significantly ameliorated, the injured area was markedly decreased, and the epithelial cell structure remained relatively intact. In addition, compared with the model group, the serum contents of total cholesterol (TC), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C) in mice of the Jinshanyu treatment groups were significantly decreased, whereas the content of high-density lipoprotein cholesterol (HDL-C) was significantly increased. Moreover, the activities of total superoxide dismutase (SOD) and reduced glutathione (GSH) in gastric tissue were notably elevated. In conclusion, Jinshanyu starch exerts a protective effect against acute gastric mucosal injury in mice by improving the oxidative stress status of gastric tissue induced by hydrochloric acid-ethanol, regulating lipid metabolism disorders, and synergistically alleviating gastric mucosal hyperemia, hemorrhage, and inflammatory damage. This finding provides a scientific basis for the development of Jinshanyu starch-based products targeting gastric mucosal injury protection.
