拟南芥中4种细胞分裂素的高效液相色谱法测定

Simultaneous Determination of Four Kinds of Cytokinin Components in Arabidopsis thaliana by High Performance Liquid Chromatography

摘要: 应用高效液相色谱法同时测定拟南芥中4种细胞分裂素组分玉米素(Z)、玉米素核苷(ZR)、6-r,r-二甲基烯丙基氨基嘌呤(2ip)和6-r,r-二甲基烯丙基氨基嘌呤核苷(2ipr)含量。结果表明,采用反相色谱柱Waters C18柱(4.6×250 mm,5 μm),在35℃以乙腈和三乙胺缓冲液为流动相梯度洗脱,流速1 ml/min,在270 nm处能准确检测出拟南芥中4种细胞分裂素组分的含量,检测限达0.001 μg/ml。

Abstract: The study was conducted to establish the methods which to detect the contents of four kinds of cytokinin components (Z, ZR, 2ip and 2ipr) in Arabidopsis thaliana simultaneously by high performance liquid chromatography (HPLC).Results showed that they could be determined precisely in Arabidopsis thaliana by using reversed phase column Waters C18(4.6×250 mm, 5 μm), at 35℃, with acetonitrile and triethylamine buffer as mobile phase, and at flow speed 1 ml/min by gradient elution and with 270 nm.The detectable limitation of this method was 0.001 μg/ml.
