摘要: 本文报道了用2,4-二硝基氟苯柱前衍生反相高效液相色谱法测定氨基酸,15种常见的氨基酸可得到很好的分离。衍生物很稳定,在室温下保存两个月无明显变化,检测极限可达到5pmol。本法可应用于食品、药品等样品的氨基酸测定。

Abstract: A method for separation and quantification of primary and secondary amino acids by pre-column derivation with 1-fluoro-2,4-dinitrobenze and reversed-phase HPLC is described. 15 amino acids can be separated thoroughly with a linear gradient elution with acetate buffer and acetonitrile solution. The products of derivation can be kept at room temperature for over two months with little change in their contents. The lowest content that can be determined was below 5 pmol. The method may be applied to the determination of amino acid composition of foods and drugs, and so on.
