    SCoT Analysis of Flowering Chinese Cabbage (Brassica rapa var. parachinensis) Based on Native Polyacrylamide Gel Electrophoresis

    In order to provide theoretical basis for the screening and identification of excellent materials and the preliminary study of resistance mechanisms in flowering Chinese cabbage(Brassica rapa var. parachinensis) cultivars, the SCoT polymorphism was used to analyze by native polyacrylamide gel electrophoresis with Caixin65, Caixin69 and F2 segregating lines. The detection efficiency, genetic diversity and differential cloning bands of native polyacrylamide gel electrophoresis were analyzed. The results showed that the native polyacrylamide gel could detect the SCoT polymorphism and genetic diversity between parents and offspring, and SCoT bands were more and clearer on native polyacrylamide gel than agarose gel, which elevated the detection efficiency of SCoT. Ten native polyacrylamide gel differential fragments were selected for cloning and sequencing, and four insect-susceptible sequences and six insect-resistant sequences were obtained. GENSCAN predicted that 8 of 10 SCoT sequences had promoter, terminal and open reading frame of genes. Homology search showed that function from 4 SCoT sequences obtained from Caixin69 included Brassica napus ubiquitin carboxyl-terminal hydrolase MINDY-1-like (LOC106367017) mRNA, B. rapa subsp. pekinensis clone KBrB026J02 complete sequence, B. rapa mitochondrial pyruvate carrier 2 (LOC103839865) mRNA, B. oleracea HDEM genome scaffold C3. Function from 6 SCoT sequences obtained from Caixin65 included B. rapa RNA pseudouridine synthase 4 mitochondrial (LOC103869454) mRNA, Trichodermaharzianum CBS 226.95 hypothetical protein (M431DRAFT_397944) mRNA, B. rapa subsp. nipposinica mitochondrial DNA complete sequence cultivar: Oushou hakusai, B. napus uncharacterized LOC106451263 (LOC106451263) mRNA, B. rapa subsp. pekinensis clone KBrB017P15 complete sequence. The analysis based on native polyacrylamide gel electrophoresis elevated the clarity and resolution SCoT marker, the detection level of genetic diversity level and accuracy of clone sequences. SCoT analysis based on native polyacrylamide gel electrophoresis was not only an efficiency method to detect genetic level, but also a simple and efficiency method to clone genes which could be used in genomics. The study improved the screening and identification efficiency of excellent strains, speeded up the breeding process, and provided a theoretical basis for further research on the mechanism of the insect-resistant breeding of flowering Chinese cabbage.

    Relationship Between Freezing Tolerance and Fatty Acid Biosynthesis in Lettuce Seeds after Low Temperature Treatment

    Hydrated lettuce seeds were the model materials for studying freezing tolerance and cryopreservation of seeds with high moisture content. Previous studies had shown that fatty acid biosynthesis pathway was involved in freezing tolerance of hydrated lettuce seeds. In this study, the moisture content of the lettuce seeds was tested after imbibition for different time. Then the difference in germination rate of seeds with different water contents after programmed cooling treatment was examined and the expression of genes involved in fatty acid synthesis (FAD2, FAD3, PPT, ELOVL), and cold response gene ICE1 were studied. The results showed that the moisture content of seeds increased with imbibition time, and the water absorbing rate showed a ‘fast-slow-fast’ trend. Cooled to the same freezing temperature (-20℃, -22℃) by programmed cooling, the seeds that imbibed less than 6 h exhibited relatively high germination rate. However, the germination rate of seeds that imbibed for more than 8 h was significantly reduced after the cooling treatment. The moisture content of imbibed seeds was saturated at 8 h, and the seeds were more sensitive in this state, which showed the effect of moisture content on freezing tolerance. After cooling treatment, gene expression was studied which indicated that the fatty acid desaturase genes (FAD2, FAD3), palmitoyl-protein thioesterase genes (PPT), and long-chain fatty acid elongase genes (ELOVL) were gradually up-regulated under freezing treatment with the extension of imbibition time for the seeds. The expression of above genes achieved the highest level when seeds imbibed for 10 h, because the hydrated seeds suffered the most serious freezing injury due to the high moisture content. Meanwhile, the expression of the antifreeze genes (ICE1) of seeds also showed an increasing tendency in imbibed cooled seeds, which achieved the highest level at 10 h. This study showed that when the moisture content of seeds was higher, the seeds would be subjected to higher freezing damage. In the same time, the seeds had a certain freezing resistance. Therefore, synthesis of more unsaturated fatty acids and antifreeze proteins by over-expression of related genes to protect the cell membrane would improve the freezing tolerance of imbibed seeds.

    Twenty-two species of garden plants commonly used in South China were selected to carry out water stress tests to investigate the physiological responses of different plants to different flooding duration in sponge city construction. The results showed that after two flooding stress tests in 25 d, the SPAD content of 22 plants decreased significantly, and the laws of soluble sugar content and starch content were different. 22 kinds of garden plants could be divided into 3 groups according to the flooding tolerance: (1) Ten species of plants with good flooding tolerance, included Cinnamomum bodinieri, Terminalia neotaliala, Nephrolepis auriculata, etc; (2) Six species sensitive tolerant plants included Osmanthus fragrans, Michelia champaca, Michelia maudiae, etc; (3) Six species of plants with normal flooding tolerant plants included Machilus breviflora, Pyrostegia venusta, Pittosporum tobira, etc.

    The DRIS Nutrient Diagnosis Analysis of N, P, K in Camellia azalea Under Different Fertilization Proportions

    To provide theoretical basis for field management and rational fertilization of Camellia azalea, the macro-elements of seedlings were diagnosed on the nursery stage. The fertilization test was carried out by the quadratic orthogonal rotational combination design with three factors using six-month-old grafted seedlings. After 12 months treatment, the concentrations of N, P and K in grafted seedlings were determined. P/N, K/N, and P/K were calculated and used as diagnostic indicators to calculate the coefficient of variance, and then evaluated the demands of N, P and K using diagnosis recommendation of integrated system (DRIS) and index method. The result showed the optimum ration of elements P/N, K/N, P/K in C. azalea was 0.074±0.108, 0.509±0.500, 0.148±0.090, respectively. The result showed system (DRIS) dynastic indicated C. azalea was established, and the response of C. azalea on N and K fertilizers was more sensitive than that on P fertilizer. Therefore, more N and K fertilizers should be applied during culture of C. azalea seedlings to promote the growth status.

    In order to explore the factors that influence the rooting of Jasminum dichotomum cutting and establish the cutting-propagation technology system, 1 year old J. dichotomum branches were used as the cutting materials. The L16(44) orthogonal experiment was conducted to evaluate the rooting behavior of cuttings from J. dichotomum as influenced by different hormone treatments, with variety, concentration, treatment time and different mediums. The orthogonal test was analyzed by using the queuing scoring method and formula scoring method and the results showed that the optimal treatment combination of the experiment was A2B3C3D2 (IBA concentration was 250 mg·L-1, NAA concentration was 500 mg·L-1, treatment time was 20 min, and substrate was sand).

    Diversity Analysis on Leaf Phenotypic Traits of the Endangered Plant Tetracentron sinense

    In order to provide scientific foundation for the conservation, development and application of Tetracentron sinense germplasm resources, 10 leaf morphological traits and 7 leaf epidermal micromorphological traits of 161 individuals in 24 populations were investigated, and then the methods such as descriptive statistics, genetic correlation analysis, principal component analysis and cluster analysis were used to discuss the phenotypic diversity of the endangered plant. The coefficient of variation among leaf phenotypic traits indexes varied from 4.21% to 49.23%, and Shannon-wiener diversity indexes varied from 0.82 to 2.07, with an average of 1.80, in which the LLEST was the maximum, and the LBPI was the minimum. Close relationship existed among leaf phenotypic traits. The contribution rate of the first three principal components reached 83.712%, which could respectively represent the distribution of phenotypic traits. Twenty-four populations of T. sinense were clustered into 3 groups when threshold was 15 in Q-clustering analysis. Results showed that the leaf phenotypic diversity of T. sinense was relatively low. Tangjiahe(TJH), Hongkou(HK) and Foping(FP) should be considered as key protection of wild populations.

    Analysis and Preliminary Selection of Plus Tree Progeny Plantation of Toona Sinensis

    An afforestation experiment of Toona sinensis containing 70 families from 12 provenances was conducted in Yongchun and Hua’an, Fujian Province. The investigation on two-year-old T. sinensis superior progeny test forest was carried. The results showed that the height and ground diameter of the progeny of 2-year-old T. sinensis in Hua’an were 1.10—2.80 m and 1.24—3.72 cm, respectively, and the height and ground diameter of the progeny of 2-year-old T. sinensis in Yongchun were 0.91—2.71 m and 1.18—3.68 cm, respectively. The difference of tree height and ground diameter between different Chinese toon families in Hua’an and Yongchun experimental sites reached a very significant level (P<0.01). The variation coefficients of tree height and ground diameter at Hua’an test site were 20.22% and 23.68%, respectively. Six families with excellent early growth performance were preliminarily selected, and their average tree height and ground diameter were 2.55 m and 3.44 cm, respectively. The average genetic gains were 23.70% and 30.16%, respectively. The variation coefficients of tree height and ground diameter at Yongchun test site were 26.44% and 26.48%, respectively. Nineteen families with excellent early growth performance were preliminarily selected. The average tree height and ground diameter were 2.52 m and 3.34 cm, respectively. The average genetic gains were 25.07% and 24.96%, respectively. Based on cluster analysis and growth comparison of height and ground diameter of superior progenies of T. sinensis, 70 families were divided into five categories: superior, superior, medium, inferior and extreme. Based on the growth performance of two experimental sites, four families, No. 4, No. 23, No. 53 and No. 65, were preliminarily identified as excellent families.

    Study on the Best Growth Model of Liriodendron chinense Plantation

    In Shimen Forest Farm of Zhenghe County, Fujian, the standard wood of Liriodendron chinense was selected for trunk analysis. Thirteen representative mathematical models such as Logistic, Webull and quadratic curve were used to measure the diameter at breast height(DBH), tree height and volume growth of L. chinense. After screening and verification, the best model for DBH growth was D=75.6887{1-e–[(T+2.3354)/19.5656]^3.1489}; the best model for tree height growth was H=(55.5498+34.2298T1.9310)/(119.6386+T1.9310); the best model for volume growth was V=2.5736{1-e–[(T–2.2455)/16.3381]^3.8741}, the determination coefficients of these three models were all 0.9999, the fitting accuracy was 99.7258%, 99.8827% and 99.7930%, respectively, and the average deviation, the average absolute deviation, the average relative deviation, the absolute value of average relative deviation were small. Compared with the measured values ??of the growth stages of each forest age segment, the data were highly consistent, the residuals were small, and the trend lines were consistent. It could be used to guide the prediction of the growth of young forests in L. chinense plantations in the northern of Fujian Province.

    The plant diversity of Lijiang National Wetland Park was reported in this paper. The results indicated that the plants were abundant, represented by 493 species, belonging to 357 genera in 132 families, and there were 165 species of wetland plants belonging to 118 genera in 54 families. In addition, the statistics and results obtained from their intensive analyses at different levels of seed plants revealed that the tropic elements were slightly higher than temperate elements, and the main geographical elements of the genera were Pantropic and Old World Tropics. Lastly, the ecological types of wetland plants were classified and counted, and the suggestions for plant protection were put forward according to the present situation.

    During the fourth survey of Chinese medicine resources and the investigation of native plants in Fujian province, six new records of plants in Fujian province were discovered, i.e. Lobelia hongiana Q. F. Wang & G. W. Hu, Lobelia alsinoides subsp. hancei (H. Hara) Lammers, Lysimachia paridiformis Franch., Oreocharis magnidens Chun ex K. Y. Pan, Pluchea sagittalis (Lam.) Cabrera, Melothria pendula L. All the vouchers specimens were preserved in Herbarium of Fujian University of Traditional Chinese Medicine.

    One genus and two species were reported as new records to Guangdong province, China. Odontochilus saprophyticus (Aver.) Ormerod., Didymoplexis pallens Griff., both of them were found in Danxia mountain National Nature Reserve. In addition, the genus Didymoplexis Griff. had not been previously recorded in Guangdong. The new records make the geographical distribution of O. saprophyticus (Aver.) Ormerod. And D. pallens Griff. more complete. Moreover, the new records provide a specific evidence of the uniqueness of climatic conditions and flora formation in Mt. Danxia.

    Based on analysis of references, network resource databases (including flora, specimen, and pictures), and also the field survey on all of the wild survival sites, the taxonomic status and morphological characteristics, places of origin and habitats, specimen records, research advances, and the endangered factors of the key national protected grade II plant Firmiana danxiaensis, were summarized in this paper, so as to providing scientific basis for its protection. There have been more than 30 years since the type specimen was collected in 1986 and named in 1987 first found in Danxia Mountain, Renhua County, Guangdong Province in China. According to the research, its areal range were enlarged from endemic to Danxia Mountain, Renhua County, to Quan’an Town and Gushi Town, Nanxiong City in 2014, and then to Yingcheng Town and Hengshitang Town, Yingde City in 2019. The habitats had enlarged from Danxia landform to limestone landform. All protection measures such as in situ and ex situ conservation, and more studies on this species should be combined for its effective protection.?

    A Bibliometric Analysis of Current Status and Trends of Domestic Horticultural Therapy

    Based on 224 Chinese documents of CNKI as of February 27, 2019, and 12 English papers published by Chinese scholars in WOS, the bibliometric analysis method was used to analyze the current status and trends of horticultural therapy in China. The study found that the research heat of horticultural therapy in China continued to rise, and the number of literatures continued to increase. However, the research network had limited branches and the research depth needed to be deepened. Medicine and health, industrial technology and agricultural science were its main research fields; landscape design and rehabilitation were the most prominent research hotspots. Finally, aiming at the limitations of domestic horticultural therapy research in terms of research strengths, funding, disciplines, publications, content and trends, some problem-focused suggestions were put forward. Furthermore, expectations for the future research were also put forward.
