Abstract:  Eucalyptus grandis is a significant fast-growing tree species. The mechanism underlying its rapid growth and its relationship with wood formation remains unclear. Glycosyltransferase GT106 family genes play a pivotal role in plant cell wall polysaccharide synthesis, yet systematic investigation into the genome-wide characteristics of GT106 genes in E. grandis remains lacking. Through genome-wide bioinformatics analysis, this study has for the first time identified 37 GT106 family members in E. grandis and comprehensively characterised their bioinformatics features. Phylogenetic analysis indicates that GT106 family genes are evolutionarily conserved among E. grandis, Arabidopsis thaliana, and Populus alba. Gene structure and conserved motif analysis showed that the EgGT106 gene contains the characteristic O-FucT plant and O-FucT like superfamily domains, with key motifs 1, 3, and 7 widely distributed. The EgGT106 promoter region is rich in light-response, hormone-response, and stress-related elements, suggesting its potential involvement in regulating E. grandis growth, development, and environmental adaptation. Furthermore, genes such as EgGT106.2, 9, and 16 were highly expressed in xylem, phloem, and leaves, whereas EgGT106.7, 11, and 23 were highly expressed in roots and stems, indicating their distinct functional differentiation. Notably, under jasmonic acid, salicylic acid, and salt stress treatments, multiple EgGT106 genes exhibited temporal expression patterns, indicating that this gene family also plays a significant role in the stress response of E. grandis. In summary, this study provides the first analysis of the characteristics of the GT106 gene family in E. grandis and the dynamics of this family under stress treatments. It establishes a theoretical foundation for further elucidating the functional roles of this family in giant eucalyptus growth and development, wood formation, and stress responses, as well as offering candidate gene resources for molecular breeding in forest trees.
