福建省枳椇种质资源遗传多样性RAPD分析

摘要: 建立和优化枳椇Hovenia acerba的RAPD反应体系,并对福建省12个种源地35份枳椇材料进行遗传多样性及亲缘关系研究。利用筛选出的10个RAPD引物共检测基因组DNA中193个位点,其中多态性位点161个(83.42%)。供试材料观测等位基因数(Na)1.8342、有效等位基因数(Ne)1.4155,Nei’s基因多样性(H)0.2493,Shannon多样性指数(I)0.3822。以遗传相似系数0.754为阈值,可将35份枳椇分成五大组。RAPD标记可有效揭示福建省枳椇种质资源的遗传多样性,所测枳椇种质资源遗传多样性丰富,极具开发利用价值。

Abstract: The methods of RAPD molecular markers were used to study genetic diversity and the genetic relationship of 35 germplasms of Hovenia acerba from 12 provenances. The reaction system of RAPD fitting for analyzing the genetic diversity of H. acerba were established and optimized. The results showed that 193 locus of genomic DNA in total were detected in 10 RAPD primers, of which there were 161 polymorphic locus (83.42%). The number of alleles (Na) was 1.8342, the effective number of alleles (Ne) was 1.4155, Nei’s gene diversity (H) was 0.2493, Shannon’s information index (I) was 0.3822. UPGMA cluster analysis showed that the similarity coefficient is about 0.754, the 35 copies of H. acerba germplasms could be classified into 5 groups. RAPD molecular markers could well reflect genetic differences and the genetic diversity among H. acerba. Thirty-five materials measured from 12 provenances of H. acerba showed abundant genetic diversity, which had great potential for exploitation and utilization.?
