基于超高效液相色谱-四极杆/静电场轨道阱高分辨质谱分析簕菜化学成分

Identification of Chemical Components of Acanthopanax trifoliatus by UPLC-Q-Orbitrap HRMS

摘要: 建立分析簕菜Acanthopanax trifoliatus中多种化学成分的超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)法,为簕菜的质量评价提供科学依据。采用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱联用仪(UPLC-Q-Orbitrap HRMS),选用Hypersil GOLD C18色谱柱(100 mm×2.1 mm,1.9 μm)进行分离,以0.1%甲酸水溶液-0.1%甲酸乙腈溶液体系进行梯度洗脱,质谱设置成负离子全扫描模式和二级质谱自动触发扫描,快速识别并通过数据库、文献对比以及对照品确认鉴定簕菜样品中的化学成分。共鉴定出29个化合物,其中19个多酚类化合物和10个萜类化合物。该研究首次报道使用UPLC-Q-Orbitrap HRMS方法快速全面分析簕菜化学成分的可行性,为簕菜活性化学成分分析提供参考。

Abstract: An ultra-high performance liquid chromatography-quadrupole/electrostatic field-orbital-trap-high resolution mass spectrometry (UPLC-Q-Orbitrap HRMS) method for the analysis of various chemical components in Acanthopanax trifoliatus was established. The sample was separated by Hypersil GOLD C18 column (100 mm × 2.1 mm, 1.9 μm), eluting with 0.1% formic acid acetonitrile-0.1% formic acid aqueous solution. Chemical components were quickly identified by mass spectrometry and comfirmed by the negative ion full scan mode and the secondary mass spectrometer automatic trigger scan. The chemical components were identified by database, literature comparison and reference materials. A total of 29 chemical components were identified, including nineteen polyphenolic compounds and ten terpenoids. This study was the first to report the feasibility of using the UPLC-Q-Orbitrap HRMS method to quickly and comprehensively analyze the active chemical components of A. trifoliatus. The method can provide a basis for the analysis of active chemical composition of A. trifoliatus.
