In vitro culture of Begonia×Manni leaves and the establishment of industrialized breeding system

丽格海棠叶片离体培养与工厂化育苗体系的建立

Abstract: The leaves of Begonia×Manni were used for in vitro culture. The results showed that B5 medium is appropriate for the induction of differentiation of leaf-derived adventitious buds. The propagation rate of adventitious buds on the B5+BA 0.5mg/L+IBA 0.05mg/L was 6.06. B5+IBA 0.5mg/L was best for rooting and the rooting rate could reached 100%, the average number of root was 4.5. The results also indicated that regulator-free medium was effective for the growth of healthy plantlet and low concentration of sucrose in culture medium played an important role in maintaining the normal morphology of plantlet. Based on the investigation, a industrialized breeding system of Begonia×Manni was established.

摘要: 利用丽格海棠叶片进行离体培养,建立工厂化育苗体系,B5培养基适合于叶片诱导分化丛芽。丛芽在B5+BA 0.5mg/L+IBA 0.05mg/L培养基中,增殖系数达6.06;B5+IBA 0.5mg/L中,生根率达100%,平均根数4.5条。生根前无生长调节剂壮苗是获得健壮植株的有效方法。培养基中附加低浓度蔗糖对维持植株正常形态起关键作用。
