Rice Transformation by Injecting Hainan Oryza meyeriana DNA and SSR Detection

海南疣粒野生稻DNA导入栽培稻及其后代的SSR验证

Abstract: Genome DNA of Hainan Oryza meyeriana with high resistance and vigorous growth was extracted, then was transformed into rice restorer R225, R128, R15 by stem injecting.From F1 generation, variation rice from others in growth period, height, type and spikelets was chosen.After 3 generation, 15 stable rice materials were screened out.The materials and controls were detected with SSR markers, which indicated that there were 4 materials from which DNA fragments specific to wild rice were amplified.The result showed that variation materials from Oryza meyeriana were acquired.

摘要: 选取抗性较好且生长旺盛的海南疣粒野生稻,提取其基因组DNA,通过穗颈注射法将其导入杂交水稻恢复系R225、R128、R15。第1代开始选择在生育期、株高、株叶型和穗粒结构等性状与群体中其他单株有明显不同的变异株,经过3代选择,筛选到15份稳定材料。利用SSR分子标记检测4份导入后代材料具有供体而受体无的DNA条带,说明本试验获得了来自疣粒野生稻的变异材料。
