Histochemical Localization of Carbohydrate and Protein during Flower Bud Differentiation in Oncidium

文心兰花芽分化过程中糖类和蛋白质的组织化学定位

Abstract: The flower buds in different development stages of Oncidium ‘milliongolds' were used as tested materials,and the changes of carbohydrate and protein during flower bud differentiation were studied by PAS method and histochemical coloration. The results showed that carbohydrate and protein mainly distributed in or around the apex of primordium. In inflorescence primordium differentiation phase, the content of carbohydrate was much less, while protein was richer and distributed uniformly. With the continued differentiation of the flower bud, carbohydrate and protein occurred abundantly in sepal primordium differentiation phase, and decreased obviously in petal primordium differentiation phase. The carbohydrate and protein focused on gynostemium and pollinia during gynostemium and pollinia differentiation phase. The contents of carbohydrate and protein in flower buds during Oncidium development process were determined, which showed that the change trends of carbohydrate and protein were consistent with the histochemical localization.

摘要: 以文心兰‘milliongolds’不同发育阶段的花芽为试材,研究糖类和蛋白质在文心兰花芽分化过程中的变化。经石蜡切片高碘酸席夫反应法(PAS法)和汞溴酚蓝组织化学染色观察发现,糖类和蛋白质主要以颗粒状分布在各器官的原基顶端及其周围。在花序原基分化期,细胞中糖含量较少,而蛋白质含量丰富且整体分布均匀。随着花芽的继续分化,糖和蛋白质含量在花萼原基分化期最为丰富,在花瓣原基时期含量明显减少;进入合蕊柱及花粉块分化期时,糖和蛋白质在合蕊柱及花粉块着生处最为集中。在文心兰花发育过程中,对不同分化时期中花芽的糖和蛋白质含量进行测定,表明糖和蛋白质含量的变化趋势与组织化学定位相一致。
