    The Preliminary Research of Antimicrobial Activity of Extract from Mahonia japonica

    Through the bacteriostatic test of extract from Mahonia japonica, one of medical plants. The result are as follows. Different substances were extracted with different extracting methods of Ultrasonic and Soxhlet in different solution and in different polar solvent. To get the antibacterial and antifungal activity by suppression of bacterial ring method and including poison of medium. The antimicrobial substance exists in the stems and leaves, and leaves is obvious; there is a little difference on extracting methods of Ultrasonic and Soxhlet. The minimal inhibitory concentration of the extract on Staphylococcus aureus, Escherichia coli, Bacillus subtilis,Micrococcus luteus, Monilia albicans are 0.6 g/mL, 1.0 g/mL, 0.8 g/mL, 1.0 g/mL, 2.0 g/mL respectively; the minimal bactericidal concentration all are 2.0 g/mL; but it is ineffective on Aspergillus niger;the optimum antibacterial effect of the solvent should be Methanol. The bacteriostatic test by different polar solvent shows that the antibacterial substance easily dissolved in nonpolar solvent such as petroleum ether, ethyl ether. The bacterial and fungal of antimicrobial experiment have a wide disparity in sensibility as follows: S. aureusB. subtilisM. luteusE. coliM. albicans,and it is no sensitive to Aspergillus niger.

    Study on the in vitro Bacteriostatic Activity of Different Solvent Extracts from Caulis Rosae laevigatae

    Caulis Rosae laevigatae was extracted with different solvents to study the bacteriostatic activity of extracts from Caulis R. laevigatae. Pseudomonas aeruginosa, Dysentery bacillus, Escherichia coli, Proteusbacillus vulgaris, Salmonella typhi, Staphylococcus aureus were used as the experimental strains. 80% ethanol Coptis extracts used as comparison. The diameters of inhibition zone were determined by agar plate diffusion method. Then the minimal inhibition concentration (MIC) were observed to test dilution methods. The results show that the water solution has no bacteriostasis function to Pseudomonas aeruginosa, Dysentery bacillus, Escherichia coli, Proteusbacillus vulgaris; The MIC to Salmonella typhi is 20 mg(crude drugs)/mL; The MIC to Staphylococcus aureus is 10 mg(crude drugs)/mL. 75% ethanol solution has no bacteriostasis function to Pseudomonas aeruginosa, Salmonella typhi, Dysentery bacillus, Escherichia coli, Proteusbacillus vulgaris. The MIC to Staphylococcus aureus is 10 mg(crude drugs)/mL. Ethyl acetate solution, chloroform solution, petroleum ether solution have no bacteriostasis function to all of the experimental strains. In Conclusion, the different solution of caulis R. laevigatae have indefinitely bacteriostasis function on common pathogenic bacteria.

    Cloning and Bioinformatic Analysis of Glucose-6-phosphate 1-dehydrogenase Gene(gsdA) from Aspergillus oryzae

    Glucose-6-phosphate 1-dehydrogenase(G6PDH) is one of the key enzymes in pentose phosphate pathway(PPP). Here, the gene encoding G6PDH is cloned from Aspergillus oryzae CICC2012. This gene was sequenced and submitted to GenBank(accession number: JN123468). The sequence was analyzed bioinformatically. The results show that G6PDH from A. oryzae is a hydrophilic enzyme consisting of 222 amino acids. The sequence from 128 to 134(DHYLGKE) is the active site, and the sequences from 170 to 176(GTEGRGG) is the possible site of coenzyme binding. The phylogenetic tree shows that G6PDH of A. oryzae is similar to other filamentous fungi and the yeast one, while distinguished from the bacterial type, the plant one, and the human one.

    The C-serum is a major component of the latex and a key material for proteinomics of Hevea brasiliensis. Using the 2-D electrophoresis, there are some disadvantages with protein extraction from C-serum, such as unsatisfied solubility, higher ions concentrations and too many impurities. In this paper, the alcohol-ether method and sonication were used to further remove interfering substances and solubilized protein based on the traditional method of TCA/acetone. Comparing the traditional method of extraction, the results showed that the new method improved the solubility, ions concentration and purity of protein samples from C-serum. The optimized methods for extraction of protein will be extremely useful for the proteomic research of C-serum form H. brasiliensis latex.

    To obtain clear Sequence-related amplified polymorphism (SRAP) fingerprints of Cymbidium, the factors influencing SRAP analysis were studied. A reliable, effective and reproductive PCR reaction system for detecting SRAP was developed. Each 30 μL PCR reaction mixture consisted of 2.2 mmol/μL of Mg2+, 0.8 mmol/L of dNTPs, 150 ng of genomic DNA, 1.5 μmol/L of primer and 2.0 unit of Taq polymerase. Samples were subjected to thermal profile for amplification in an oven thermocycler: 4 min of denaturing at 94 ℃, five cycles of three steps, 1 min of denaturing at 94 ℃, 1 min of annealing at 35 ℃ and 1 min of elongation at 72 ℃, in the following 30 cycles the annealing temperature was increased to 55 ℃, with a final elongation step of 5 min at 72 ℃.

    Mesophyll protoplasts were isolated and purified from leaves of in vitro propagated Polgostemon cablin (Blanco) Benth. cv. shipaiensis, and different factors affecting protoplasts yield and activity were investigated. The results showed that young leaves of seedling subcultured for 12~22 days in the enzyme solution consisting of 0.5% (w/v) pectolyase Y-23, 0.2%(w/v) Macerozyme R-10 and 1. 5% (w/v) cellulase R-10, in 11% (w/v) mannitol buffered with 0.1% MES and 0.02% CaCl2 for 8 h, the yield was 1.85×107 protoplasts per gram leaves (fresh weight), the viability was above 89%.

    Effects of Paclobutrazol and ABA on Endogenous Phytohormones and Photosynthetic Rate of Michelia alba

    Michelia alba, with a high height and few flowers in blooming period, is not good for cultivation in house. To overcome these disadvantages, Michelia alba were treated with paclobutrazol and ABA in various ways, such as soil application, foliage spraying, and used to detect the height, blossom condition, phytohormones concentrations and photosynthetic efficiency in different positions of plant. The results showed that the concentration of phytohormones, such as IAA, GA3, Z, ZR and ABA,displayed different regularity of change, and the photosynthesis rate changed obviously after treatments. The plants treated was short and small, flowering early, and the amount of flowers and flowering frequency increased. Soil application and foliage spraying of paclobutrazol and ABA were found the best way to dwarf the height of Michelia alba to 40%~50% of CK, and increase the amount of flowers 2.3 times of CK.

    Effects of Mn2+-Fe2+ Combined Stress on Physiological Indexes of Rivina humilis

    The effects of Mn2+-Fe2+ combined stress on physiological indexes of Rivina humilis was studied by the method of nutrient solution culture. The results showed that both the chlorophyll content and the soluble sugar content were significantly higher than those of the control and reached the maximum when R. humilis were stressed by 6 mmol/L Mn2+ and 0.5 mmol/L Fe2+. At the same time, the MDA content was extremely significantly lower than that of the control and got to the minimum. Additionally, the combined treatment of Mn2+-Fe2+ also improved the activities of SOD, POD and CAT. In a word, the suitable combined concentrations of Mn2+ and Fe2+could alleviate the toxicity of single metal to some extent, and enhanced the tolerance of R. humilis to the ferromanganese stress.

    Wild Aromatic Plant Resources and Their Exploitation and Utilization in Jiufu Mountain, Fujian Province

    The wild aromatic plants resources were investigated in Jiufu Mountain Nature Reserve in Youxi, Fujian province. The results show that there are 132 species of wild aromatic plants, belonging to 91 genera in 47 families in this area, which accounted for 52.8%, 29.7% and 17.4% in families, genera, and species of the wild aromatic plants in Fujian province respectively. Among these aromatic plants, Lauraceae is the biggest population, including 22 species. Other advantage families, both Rosaceae and Orchidaceae include 10 species. Lamiaceae includes 9 species and Asteraceae includes 7 species. Magnoliaceae and Oleaceae include 5 species, 4 species respectively. The smaller population were Rutaceae, Primulaceae, Moraceae, Hamamelidaceae, including 3 species. The dominant genera are Lindera (6), Cinnamomum(4), Machilus(4), Rosa(4), Artemisia(4), Litsea(4), Photinia(3), Lysimachia(3). The wild aromatic plants with greater value of development and utilization are Pinus massoniana, Sarcandra gladra, Houttuyia Cordata, Litsea cubeba, Cinnamomum camphora, Machilus grijsii, Machilus pauhoi, Litsea aggregata, Litsea elongat, Elaeagnus pungens, Mosla punctulata, Liquidamber formosana, Rosa laevigata, Gardenia jasminoides, Lonicera nitida, Artemisia argyi and so on. The species composition, aromatic position, live form, distribution and the suggestions are put forward on their development and utilization.

    In this article, basis on the investigation and previous researches on kiwifruit resources in Mount. Emei, the species diversity and geographical distribution of kiwifruit in Emei Mountain were reviewed. Furthermore, the reason that there were so many varieties and species in Emei Mountain was discussed. Those results may contribute to studies on kiwifruit growing conditions and environmental factors, and accelerate the process of domestication and cultivation of kiwifruit.

    Community Characteristics of Endangered Species Rosa praelucens

    The community structure of Rosa praelucens were studied by using the sample plot method .The results show that:(1) There were 97 vascular plant species in the community belonging to 24 families and 71 genera. And, there were 6 fern plants species belonging to 2 families and 5 genera, gymnosperms have 1 species belong to 1 family. There were 90 species of angiosperm plants belonging to 65 genera and 21 families. (2) The community structure was complicated which could be deviled into tree layer, shrub layer, herb layer, and less interstater plants. Herb layer grew better than tree layer and shrub layer. According to Raunchier, the high bud plant has high percent of 39.56% in all plants.

    An Investigation of Wild Fruit Plants Resources in Dongyuan Kanghe Provincial Nature Reserve, Guangdong Province

    According to the investigation and correlative data statistics, we find out that in Dongyuan Kanghe Nature Reserve (KNR) in Guangdong Province, there are 70 species wild fruit plants in all, which belong to 27 families and 45 genera. The species of wild fruit in KNR account 6.94% for the KNR total vascular plants (1 023 species), about 30.34% for the wild fruit (234 species) of Guangdong Province, or 6.41% for the National wild fruit (1 076 species). Therefore, KNR has rich fruit resources, including dicotyledonous of 24 families, 43 genera and 67 species, and monocot plants section of 2 family, 2 genera and 4 species; among of them, there are 20 species of trees, 36 species of shrubs, vines 13 species and herbaceous 2 species; berry 53, drupes 10, pome 6, hesperidium 2; 16 tropical families, accounting for 59.26% and 7 temperate distribution families, accounting for 25.93%, so the tropical distribution takes the advantage. Sixty-two species can be used for medicinal purposes, decorative plant 35, the timber wood 17, the textile fiber 12, the tanning material plant 9 and so on. Most of the wild fruits in KNR have strong vitality, and can be eaten by raw for unique taste, high nutritional value and so on. Based on the present situation of wild fruit plants in KNR, the authors put forward some related suggestions.

    According to the investigation on the vegetation in Fujian Province, 4 Newly recorded genera, Galactia, Gueldenstaedtia, Mercurialis, Eryngium, and 5 newly recorded species, Galactia tenuiflora, Gueldenstaedtia verna, Mercurialis leiocarpa, Herissantia crispa, Eryngium foetidum of Angiosperm were found. The voucher specimens were deposited in the Herbarium of FJTCM.

    The soil water-holding capacity of ten transformed plantations at Lishui, Xi’an, Xijiaoshan in Nanhai district, and Sanzhou in Gaoming district of Foshan City were studied. The results showed that there were differences in soil physical characteristic among different transformed stands. The analysis found some significant correlation between some soil physical factors. Ten transformed stands were divided into four types by Cluster analysis.

    The Effect of the Characteristics of Cinnamomum camphora var. linaloolifera Cuttings on Their Survival Rate

    Cinnamomum camphora var. linaloolifera is an elite variety of oil use forest selected from C. camphora germ plasm resources, and it can maintain excellent economic traits only to adopt vegetative propagation. Using Greenhouse for containerized cutting seedling experiment, the results show that the character of C. camphora var. linaloolifera affect on the cutting survival rate was significant or extremely significant. The survival rate of the top significantly higher than the middle, the lower and the base in the same branch; the strip than 10 cm long, 0.3 cm diameter, with two and a half leaf, a slope cut shape, cutting survival rate higher than other treatment. The optimal combination is the closed shoot tip 1~2 section in the same shoot, 10 cm long, diameter of about 0.3 cm, with two and a half leaf, the shape of below cut is slope.

    Using stem sections of Philodendron congo which was introduced recently as explants, the study on In vitro rapid propagation of P. congo was carried out in this test. The results showed that MS + 6-BA 2 mg/L + NAA 0.1 mg/L was much better medium for bud multiplication, and the best medium for rooting was MS + IBA 0.5 mg/L.

    Cycads are the most primitive living seed-plants. Studying on the pollination biology of cycads has significant value for researching into the origin, phylogeny of seed plants, the evolution of dependent mutualism between the insects and cycads, as well as the mechanism of reproduction and the mechanism of endanger. In this review, we summarize the information available concerning the pollination biology of cycad, including morphology and structure of male and female cone, floral phenology of male and female plants, the medium of pollination and the mechanism of pollination. For future research, we suggested that more studies should be carried out on the pollination of cycad (especially for the Cycas) in a wider range, and the exclusion method should be conducted to determine the medium of pollination of cycad more widely, and more detailed studies should be done about the problem of that insect is in highly specialized dependent mutualisms with cycads.
