    Establishment of a High Efficient Gene Editing System for Nicotiana attenuata

    Nicotiana attenuata is a model plant species for study of plant-insect and plant-pathogen interactions in the genus of Nicotiana. In order to provide a more powerful and efficient means to study gene function in N. attenuata, we explored and finally established the highest gene editing efficiency system for N. attenuata by using phytoene desaturase gene (PDS) as a target gene, pHSE401 as the multi-target gene editing vector, and hygromycin as the selecting marker. Finally, we obtained a gene editing efficiency of approximately 80% for the PDS gene, far exceeding the approximately 30% gene editing efficiency currently reported in N. attenuata. The gene editing efficiency was also validated by targeting the WRKY70 gene. After sequencing, it was found that the editing frequency of WRKY70 was 83%, with 50% large fragment deletion. Taken together, we successfully established and verified a gene editing system with the highest editing efficiency reported ever in N. attenuata, thus providing a solid foundation for future gene function research in N. attenuata.

    Differential Zymogram Pattern of Protease in Leaves of Brassica napus Seedlings under Low Phosphorus

    To investigate the effect of low phosphorus stress on the expression of protease zymogram content in seedling leaves of Brassica napus, Dingyouza No. 4 was used as experimental material. The seedlings were subjected to phosphorus deficiency nutrient solution for 10 days under low phosphorus stress, and leaf proteins were extracted by direct grinding method. Optimized non-denaturation two-dimensional electrophoresis combined with protease detection technology was used to obtain a good protease separation map. Protein difference analysis was performed using ImageMaster 2D Platinum 7.0 software. The results showed that the expression levels of 6 protease points in rape leaves were significantly different under low P stress (the difference was more than 2 times), among which 3 were significantly up-regulated and 3 were significantly down-regulated. The results suggested that different leaf proteases are involved in the response pathway of rape to low phosphorus. The two-dimensional zymogram technology based on non-denaturing two-dimensional electrophoresis mentioned in this paper also laid a foundation for the study of protease zymogram.

    Released Characteristics of Volatile Organic Compounds in Cunninghamia lanceolata

    To clarify the composition and variation rules of contents of components of volatile organic compounds (VOCs) in Cunninghamia lanceolata, seasonal changes of composition and relative contents of components of VOCs released by C. lanceolata were compared and analyzed by using dynamic headspace air-circulation method and thermal desorption-gas chromatography-mass spectrum (TDS-GC-MS) technique. The results showed that total of 60 compounds were identified in the VOCs released by C. lanceolata. The number of compound types rose first and then decline in all year, and the number of compounds released in summer was the most abundant, reaching 42. The largest number of detected compound types consisted of alkanes, aldehydes, esters, and alcohols, accounting for 73.33% of all types of detected compounds. Compound types of C. lanceolata VOCs released in Spring were mainly alcohols, with the total relative content of 27.19%, terpenes in Summer, with the total relative content of 17.29%, and aldehydes in Autumn, with the total relative content of 23.55%. In summary, C. lanceolata has a certain intensive effect on health care, and summer is the best time for forest recreation in C. lanceolata forest.

    Screening Excellent Pollen Parents for Hybridization of Camellia azalea as Seed Parent

    In this research, Camellia azalea was used as seed parent and 13 cultivars of Camellia were used as pollen parents. A total of 13 cross combinations were designed by the conventional methods of crossbreeding. The fruit setting rate, seed setting ratio, emergence rate and single flower seedling rate were used as indexes to evaluate the affinity and the results were obtained. Factor analysis method was used to comprehensively evaluate the affinity of hybrid combinations. The results showed that?‘Ludaikena’, ‘Mali feisi’, ‘Nilifuren’, ‘Hualuzhen’ and ‘Walinasheng’ were screened out with high affinity for hybridization as pollen parents and C. azalea × ‘Ludaikena’, C. azalea × ‘Malifeisi’ combinations with high affinity were selected. The characteristics of flower color and flower form in C. azalea F1 depend on the characteristics of pollen parent, the leaves more similar to the female parents, and the hybrid offspring inherited the characteristics of year-round flowering of the female parents.

    Comprehensive Evaluation on Picking Efficiency of Camellia oleifera Clones Fruits Based on Their Canopy

    Ten clones of Camellia oleifera were used as tested materials, the differences in fruit traits and fruit picking characteristics were analyzed and compared between outside and inside of canopy, and were comprehensively evaluated by principal component analysis and cluster analysis. The results showed that yield per m2 and fresh fruit weight per plant of Changlin4 and Changlin53 were higher inside crown than those of outside crown, fruits picking efficiency of Changlin3, Changlin4, Eyou81, Eyou102 and Eyou151 outside crown were more than inside crown. Fresh fruit quality per plant, yield per m2, picking efficiency of Eyou81 were higher than those of other clones, which were 9.88 kg, 3.94 kg·m–2, 7.96 g·s–1 respectively, followed by Changlin53, Changlin4, Changlin40, Eyou102 and Eyou63. Principal component analysis was used to summarize the 17 indicators into four principal components. The comprehensive ranking of 10 clones was as follows: Eyou81> Changlin53> Changlin4> Changlin40> Eyou102> Eyou63> Changlin23> Changlin18> Changlin3> Eyou151. Cluster analysis divided 10 clones into three categories, which were high yield and efficient harvest varieties, low yield and low efficiency harvest varieties, and medium-excellent varieties. The research results could provide a basis for lightly simplified improved variety the screening of C. oleifera.

    Bryophyte Diversity and Environmental Response in University Campuses in Karst Mountainous Regions:

    In this paper, the species diversity and habitat characteristics of bryophytes in universities in Karst mountains were studied, and the relationship between bryophyte growth and environment was discussed. The results showed that there were 51 species of bryophytes belonging to 27 genera in 12 families. The dominant families were Pottiaceae, Brachytheciaceae and Hypnaceae. The main life forms were clustered, interlaced and cushion. The order of biodiversity in different function zones from high to low was administrative office area, leisure area, teaching and scientific research area, sports area, dormitory rest area. The order of species richness in different habitats from high to low was open forest lawn, underforest, lawn environment, shrub pond, road. The degree of human disturbance, light and humidity were important factors that change the diversity composition of bryophytes. In conclusion, the bryophyte diversity in Guizhou University was low, and most bryophytes were native mosses. In the microhabitat of moss growth, woodland and open forest lawn with low disturbance and high air humidity were more conducive to moss growth.

    Based on the fourth national survey of traditional Chinese medicine resources, the species and distribution of medicinal plant resources in Anxi were analyzed by means of field investigation, market investigation, visiting investigation and consulting relevant data. The results showed that there were 1515 species of medicinal plants in Anxi, belonging to 215 families and 844 genera. Herb was the main living type, and the medicinal parts were mainly roots, rhizome and whole grass. There were 115 kinds of important medicinal materials and 46 kinds of characteristic medicinal materials in Fujian. Anxi was rich in medicinal plant resources and had high species diversity. In the future, we should strengthen the protection of resources and rationally develop and utilize the wild plant resources in Anxi.

    The paper reported two newly recorded genera, Stimpsonia Wright ex A. Gray and Chikusichloa Koidz., and three species, S. chamaedryoides Wright ex A. Gray, C. aquatica Koidz. and Passiflora kwangtungensis Merr., from Guizhou Province. The voucher specimens were collected from Taipingshan Prefucture Nature Reserve located in Liping County, Guizhou Province, and deposited in the herbaria of South China Botanical Garden, Chinese Academy of Sciences (IBSC) and Institute of Botany, Chinese Academy of Sciences (PE). The brief descriptions of the three plants were provided. The variation on leaf blades of P. kwangtungensis was also discussed.

    In order to establish an efficient and practical tissue culture and rapid propagation system for Bletilla striata, the mature and undehisced capsule, tuber and leaf were used as explants to screen suitable explants, and the effects of medium with different growth regulator concentrations on the germination of protocorms, the induction of clustered buds and the rooting were studied. The results showed that seed was the best explants material. The medium with KC + NAA 0.5 mg·L–1 was beneficial to cluster bud formation from protocorms. The medium with MS + 2.0 mg·L–1 6-BA + 0.2 mg·L–1 NAA + 10% coconut juice was beneficial to proliferation and seedling formation of multiple shoots. The best rooting medium was 1/2MS + NAA 0.5 mg·L–1 for transfering 2 cm height buds, and the rooting rate was 98.67%.
