    Study on Some New Flavonoid C-glycosides and Isomers of Flavonoids from Lotus Plumule Using Liquid Chromatography-Mass Spectrometry

    In this study, electrospray ionization mass spectrometry (ESI-MS) was used to analyze flavonoid C-glycosides isolated from lotus plumule. It was found that in negative ionization mode, flavonoid C-glycosides with hexose substitution experienced cross-ring cleavages of sugar moiety, characterized by loss of a series of neutral fragments (90 u, 120 u, 150 u) which distinguished markedly with flavonoid O-glycosides. Flavonoid O-glycosides with one hexose substitution lost sugar moiety (162 u) directly. Although the types of hexose could not be directly differentiated by MS method, it could be solved by means of liquid chromatography-mass spectrometry (LC-MS) due to their different retention times. For flavonoid O-diglycosides, main fragment ions were derived from the loss of sugar moiety (146 u, 162 u, 308 u) and the types and the linkage information of disaccharide could be differentiated by the fragment ions and its relative abundance in the MS spectra. Not only can differential MS fragmentation patterns of flavonoid C-glycosides and flavonoid O-glycosides be very helpful to the rapid identification of flavonoids from lotus plumule, but can also enable the better differentiation of isomers of flavonoids from lotus plumule.

    The optimum extraction conditions of tenacissima G from Marsdenia tenacissima were investigated. On the basis of single factor test, the alcohol concentration, solid-liquid ratio, extraction duration and extraction times were selected for the four-factor, three-level Box-Behnken experimental design. In addition, Design Expect 8.0 software was employed to analyze the test data and the response surface methodology was applied to optimize the extraction conditions. The results suggested that optimum extraction conditions consisted of 80% ethanol as the extraction solvent, an extraction duration of 1.5 h, a solid-liquid ratio of 1∶20(W/V) and extraction of two times. Under these optimum conditions, the extraction yields of tenacissima G from M. tenacissima was 0.253%.

    To study the antioxidant activity of the ethanol extraction of Pyrrosia petiolosa, DPPH radical scavenging, reducing power, total antioxidant activity and hydroxyl radical scavenging assay were determined. The results showed that the IC50 of the DPPH· and ·OH radical scavenging were 75.82 and 46.30 μg·mL-1, respectively. Compared with the ascorbic acid, the ability of DPPH free radical scavenging was higher, while the ·OH radical scavenging activity was lower. It indicated that the ethanol extraction of P. petiolosa had strong antioxidant activity.

    Optimization of Extraction Process of Total Flavonoids from Lamiophlomis rotata

    Taken the extraction rate of total flavonoids from Lamiophlomis rotata as index, extraction methods, the alcohol concentration, solid-liquid ratio, extraction duration and extraction times were compared. Based on signal factor results, orthogonal test factors and levels were determined to optimize extraction condition by L9(34). The optimum extraction process were as follows: extraction solvent was 80% ethanol, solid-liquid ratio 1∶50, extraction time was 25 min for 3 times. The extraction process was simple and reasonable, which could effectively improve the extraction rate of total flavonoids from L. rotata.

    Effects of Heavy Metal Ions Stress on Seed Germination of Sesamum indicum and Its Seedling Growth

    The effects of Cd2+, Ni2+, Cr6+, Hg2+ and Pb2+ on sesame (Sesamum indicum) seed germination and seedling growth were researched. The results showed that heavy metal ions stresses had less influence on sesame seed germination and the most influence on radicel growth. With increasing concentration of heavy metal ions, sesame seedling shoot length, root length and seedling fresh weight decreased continuously. After seed germination, sesame radicle could not grow and seedlings did not survived with the concentration of Cd2+, Ni2+, Cr6+, Hg2+ or Pb2+ up to 20, 20, 20, 50 or 100 mg稬-1, respectively, which illustrated these concentrations could be used as lethal concentrations of sesame seed germination. By comparison, the toxicity degree of 5 kinds of heavy metal ions on sesame seed germination and seedling growth was Ni2+ > Cr6+ > Cd2+ > Hg2+ > Pb2+.

    Physiological and Biochemical Influences of 6-BA and GA3 on Red-stem Dendrobium officinale under Salt Stress

    Physiological and biochemical influences of plant growth regulators(6-BA and GA3) on red-stem Dendrobium officinale seedlings under salt stress were performed. The results showed that low concentration of 6-BA and GA3 remarkably increased stem growth rate of red-stem D. officinale under salt stress, but reduced root growth rate. And GA3 had more effects than 6-BA in root growth rate. With the increase of 6-BA concentration, the chlorophyll contents in leaves of red-stem D. officinale displayed a high-low trend, and the carotenoid contents displayed a low-high-low trend, while high concentration of GA3 increased the contents of chlorophyll and carotenoid; Low concentration of 6-BA increased the content of soluble sugar in leaves of red-stem D. officinale under salt stress, while other concentrations of 6-BA and GA3 reduced the contents of soluble sugar. The results revealed that treatments of appropriate concentrations of exogenous plant hormones can protect the seedlings of red-stem D. officinale from hurt caused by salt stress by improving the photosynthetic pigment synthesis and soluble sugar accumulation.

    Changes in Levels of Endogenous Plant Hormones During in vitro Rapid Propagation of Tripterygium wilfordii

    The dynamic changes of endogenous plant hormones from in vitro seedlings of Tripterygium wilfordii were determined. The results showed that the culture of 7—14 d is the key stage of bud induction. It has been basically completed in 21 d. Lower concentrations of GA3 is conductive to the growth and differentiation of buds. In the training of 28—35 d, the bud growth began to enter the stage of slow inhibition; in the process of bud subculture, we should properly control the culture time, timely transfer, reduce the content of the auxin and increase the content of auxin. At the same time, high ratio of GA3/IAA and low ratio of ABA/IAA is good for the bud differentiation and proliferation of T. wilfordii; in seedling cultivation process, higher levels of auxin and gibberellin were needed. After the culture of 14 d, the medium should be replaced in time. In the culture of 7th day, the roots begin to grow. In the early stage of root formation, low concentration of ABA is favorable for the formation of callus at the cut point. At the early stage of root development, the low ratio of ZT/IAA and high ratio of GA3/ABA were favorable to the rooting culture of T. wilfordii.

    The contents of N, P, K, Ca and Mg in the leaves of seedlings of 4 tree species were analyzed by semi-micro kjeldahl method, molybdenum blue colorimetry and atomic absorption spectrophotometry, respectively. The results showed that contents of Ca and N were high, whereas contents of K, P were low in Cinnamomum camphora seedlings; the contents of K, Ca and Mg were low in Elaeocarpus sylvestris seedlings; the contents of N, K and Mg in Schima superba were the highest among the seedling of 4 tree species; P content was the highest and N content was the lowest in Syzygium cumini seedlings. Growth characteristics of C. camphora and S. superba were seedlings with low seedling height and large crown, whereas those of E. sylvestris and S. cumini were seedlings with high seedling height and small crown.

    The buds from ‘Huanghua’ and ‘Mixue’, two cultivars of Pyrus pyrifolia, were used as the experiment materials to explore the characteristics and expression of dormancy-associated MADS-box (DAM) genes PpMADS13-1 and PpMADS13-3 in different chilling requirement pear cultivars from Fujian. PpMADS13 genes were cloned and used the QRT-PCR to analysis their expression changes during the dormancy period. The results showed that PpMADS13-1 was conservative in low chilling requirement pears. The conservatism of PpMADS13-3 was not so high in different chilling requirement pears. The PpMADS13-1hh and PpMADS13-3hh in ‘Huanghua’ were gradual down-regulation with dormancy release. The PpMADS13-1mx and PpMADS13-3mx in ‘Mixue’ presented a trend of ‘M’ type during the dormancy. The result indicated that both PpMADS13-1 and PpMADS13-3 work in dormancy release, but their regulation model were different between two pear cultivars.

    To explore the transcriptome during floral organ development in Asian lotus(Nelumbo nucifera), the total RNA of flower buds was isolated from three typical lotus cultivars: N. nucifera ‘Honghu Honglian’(single flower), N. nucifera ‘Tangzhaotisi Lian’(double flower) and N. nucifera ‘Qianban Lian’(fully-doubled and thousand-petalled). The double-stranded cDNA was synthesized based on the extracted mRNA using SMART method, then digested with restriction enzyme SfiI, and finally purified and ligated to the pUC19 vector. Thereafter, the flower buds cDNA library was constructed and amplified. The constructed cDNA library had the titer of 1.12×106 pfu·mL-1. The insert sizes of the library were 500—2000 bp and the recombination efficiency of cDNA library was about 95%. The cDNA library would provide the fundamental information for studying on gene expressions and regulations during floral organ development in lotus flower buds.

    In this study, total RNA extraction of roots, stems, and leaves from Cinnamomum kanehirae was carried out. Electrophoresis, Nanodrop and Aglient 2100 analysis indicated that the value of 28S/18S was 1.7 to 2.1, RNA?integrity number was higher than 7, and RNA concentration was higher than 50 ng·μL-1, indicating that RNA was intact. In summary, the study showed that the purity, concentration and?integrity of the isolated RNA were qualified, and could be used for subsequent molecular biology studies.

    The species association of 20 dominant species of Castanopsis community in Jiulianshan Nature Reserve was analyzed by variance analysis, χ2 test, association coefficient(AC) and jaccard index(JI). It was found that the overall association between 20 dominant species was not significantly positive. The χ2 test showed that among these 20 dominant species, 3 species pairs exhibited significant positive association, 1 species pairs exhibited highly significant positive association, 1 species pairs exhibited significant negative association, 3 species pairs exhibited highly significant negative association. Most of the total 190 species pairs did not show significant association. The number of significant negative association between high important value species was most, the number of significant negative association between high important value species and low important value species was neutral, the number of significant negative association between low important value species was least. The community was in the phase of succession toward the stable stage.

    The niche characteristics of 9 tree species of Phoebe chekiangensis community in Sanqingshan Mountain were analyzed. The niche breadths and niche overlaps of main species in the community in the sample plot were measured by using Shannon-Wiener index and Hurlbert index. The results showed that the niche breadth of Phyllostachys heterocycla, Liquidambar formosana, Trachcarpus fortunei and Phoebe chekiangensis were bigger,which of Pterocarya stenoptera, Camellia oleifera, Castanopsis sclerophylla, Armeniaca mume and Cyclobalanopsis glauca were smaller. The niche overlap of Cyclobalanopsis glauca and Castanopsis sclerophylla was the biggest among all of species, Phyllostachys heterocycla or Liquidambar formosana and Armeniaca mume was the least. The wider of niche breadth, the higher capability of adapting environment, the more powerful capability of utilization resources. The niche overlap among species was higher, the niche similarity between species was greater, similarity of utilization resources was more powerful. The niche overlap between species with broad niche breadth was generally greater, which between species with narrow niche breadth was greater in the same or similar environment requirements, which between species with broad and narrow niche breadth was smaller.

    Investigation on Medicinal Plant Resources in Huazhou City in Guangdong Province

    To understand the present status of medicinal plant resources in Huazhou in Guangdong province, diversified investigation methods including wildlife appraisal, sample collection and evaluation were carried out.燭he results showed that there were 99 families including 338 species medicinal plants in Huazhou; 43 species of them were key breeds, and 19 species of them were special breeds Chinese medicines in Guangdong province. In conclusion, there was an average pool of resources for medicinal plants in Huazhou, the wildlife population that could be served as potential candidates for medicinal usage had been severely damaged, sufficient methods currently were utilised for cultivation of local medicines. Exploitation and utilization medicinal plant resources, protection existing species should be emphasized.

    Biodiversity of Landscape Trees in Fuzhou Housing Estates

    This study evaluated species composition and floristic diversity of landscape trees in 36 housing estates in Fuzhou. There were 12 694 trees belonged to 155 species, 97 genera and 42 families. The species were divided into 6 frequency groups. 20 species with 7560 trees were in the Signature group. The Dominant, Common, Occasional, Rare and Solitary groups contained 17, 19, 49, 38 and 12 species with 2383, 1297, 1269, 173 and 12 trees respectively. The landscape trees in Fuzhou showed lower biodiversity indexes, especially species richness, compared to developed cities. The landscape trees in aged estates and relocation estates were with lower biodiversity than those in other new estates.

    Through wild plant resources investigation and specimen collection in Fujian province, four new distribution records of seed plants—Paris fargesii Franch., Stellaria wushanensis Williams, Chrysosplenium delavayi Franch. and Isoglossa collina (T. Anders.) B. Hansen were found. The voucher specimens were preserved in the herbarium of Fujian Agriculture and Forestry University(FAFU).

    Effects of fertilizers with different N-P-K ratios and applying methods on growth of Dendrobium nobile tissue cultured seedling were studied taking tissue cultured seedlings of Den. Lucky Girl ‘Sweetheart’ as materials. The results showed that treatments of foliar spraying and root irrigation with peters No.2 10-30-20 had the highest survival rates. Plant height, leaf number and stem circumference had moderate performance with the treatment of root irrigation with peters No.1 20-20-20, but the bud number was the fewest. The treatment of root irrigation with peters No.10 30-10-10 had the best performance in the vegetative growth, but the lowest survival rate. The treatment of root irrigation with 10-30-20 had the most shoot number, but the worst performance in the other indicators.

    The cutting rooting of Camellia edithae under different plant growth regulators NAA, IBA and ABT-1 were carried out. The results showed that the suitable rooting condition was 500 mg·L-1 NAA for 20 minutes, the rooting rate could be up to 93.33%. And the suitable matrix was yellow soil and river sand with ratio of 3∶1.

    Sponge gourd cultivar ‘Fuyan 1’ was F1 from ‘SG-9’ and‘SG-17’, displayed early-middle ripe character,with more high rate of setting fruit. The fruit was short stick, non-crack, green skin with obvious black stripes, and weighed about 510 g with 20—25 cm in length, 5—7 cm in diameter. The flesh tasted sweet and hard, be difficult to get brown. The yield was 45 000—60 000 kg·hm-2, and was suitable to be planted in Fujian province and periphery provinces.

    Eleutherococcus trifoliatus belongs to Araliaceae family, having high edible and medicinal value. Thus the attention to E. trifoliatus has been grown. The chemical compositions of E. trifoliatus are mainly flavonoids, caffeoyl quinic acid tannins, polysaccharides, saponins, volatile oil, etc. Studies on pharmacological and toxicological activity showed that the total flavonoids from E. trifoliatus had anti-inflammatory effect, antioxidant, antibacterial antiviral effect and so on, and it had no acute toxicity and genetic toxicity. E. trifoliatus has been developed as many products for example, anti-inflammatory spray, toothpaste, Bai-Le-compound, transparent soap, tea, etc. The researches on chemical constituents and pharmacological activities in E. trifoliatus were summarized, and its related health products research and functional products were described in this review. It was suggested that E. trifoliatus could be used as important food and medicine homologous material. However, the study of its components was not comprehensive enough. This review would be useful for deeply development of E. trifoliatus as nutraceutical material.

    The virus disease is one of the greatest threats for the cultivation of peppers. In particular, tobacco mosaic virus (TMV) which is the first described virus in detail is a primary destructive pathogen that targets Solanaceae crops, especially pepper and tomato. It is essential to definite the mechanism of resistance to TMV and identify the resistance genes and germplasm. Capsicum plants have genes, designated L genes, providing resistance against TMV.
