    Cloning Vernalization-Related Gene VRN2 and Construction of its Plant Expression Vector in Arabidopsis thaliana

    In this paper, the vernalization-related Gene VRN2 cDNA from RNA of Arabidopsis thaliana L., GeneBank accession number AY063047 was successfully cloned.The cDNA sequence of VRN2 was 1 354 bp and its region in encoding was 1323 bp.The protein deduced from VRN2 contained 441 amino acids.The cloned cDNA of VRN2 was introduced into a middle vector pBPFΩ7.The fragment with P35S promoter and nos were digested by PstI, which were connected to pBI121 vector, giving plant espression vector containing VRN2 gene.

    The effects of different substrates and growth regulators on photosynthesis and water use efficiency (WUE) of Artocarpus lingnanensis were studied in this paper.The results showed that net photosynthetic rate (Pn), transpiration rate (Tr), WUE of A.lingnanensis cultivated in improvement substrate (S2) with appropriate IBA and NAA were enhanced.The assimilation products of A. lingnanensis were remarkable enhanced in S2 compared with in S1, and Pn, stomatal conductance (Gs), Tr, WUE and chlorophyll b (CHLb) were also significant raised;Pn, WUE, total chlorophyll (TCHL), chlorophyll a (CHLa), ChLb of treatment with appropriate IBA and NAA were significantly higher than that of CK and IBA treatment.The results indicated that S2 with appropriate IBA and NAA was the best treatment of to improve photosynthesis and assimilation of A. lingnanensis.

    Fern is an old fibrovascular plant and also a unique species in the evolution of botany system.The active change of the protective enzyme of Polystichum neolobatum and Dryopteris fuscipes under the compound stress of different Cd-Pb concentrations was studied in this article.It was discovered that the activeness of Polystichum neolobatum's protective enzymes, POD, SOD and CAT, had a positive collaboration with the given concentration.In other words, with the increasing of given concentration, the activeness of enzyme also increased evidently.The activeness of Dryopteris fuscipes' protective enzymes, POD, SOD, CAT, was repressed in the given concentration of 600+1200mg/kg, while was promoted in 200+400mg/kg and 400+800 mg/kg.

    Application of thermal diffusion technology to measure the sap flow of Pinus massoniana in serious soil erosion areas in Changting county during the typhoon, the results showed that:typhoon brought high-intensity rainfall and strong winds which suppressed transpiration;the rainfall didn't improve plant availability moisture;photosynthesis and water physiology before and after the typhoon had no significant difference;the sap flow density during the typhoon was significantly related (P<0.01) with the environmental factors as:solar radiation, air temperature and vapor pressure deficit, while on sunny days were:air temperature, solar radiation, relative humidity and vapor pressure deficit.Studies have shown that:typhoon did not improve the plant available water, nor destroy the physiological activities of Pinus massoniana.

    In terms of tree type, tree height, diameter at breast height, crown width, stem strength, wood density, foliage layer and root morphology, which were all related with the wind-resistance, graduation and comprehensive evaluation of tree species wind-resistance were studied by comprehensive evaluation and grey relational grade method in this paper.For 82 tree species in Zhanjiang city, the classification result was that:They were classified into four classes including class Ⅰ with 19 species, class Ⅱ with 20 species, class Ⅲ with 31 species and class Ⅳ with 12 species, and grey relational grade was much fitter to evaluate and grade the wind-resistance of tree species than comprehensive evaluation.

    According to the study on carbon sequestration of vegetation in Eco-economic Zone of Poyang Lake, the total carbon storage of forest is 364.3×106(t), the carbon sequestration of grassland vegetation is 90.65×104 (t), the carbon sequestration of rice is 18.51×106 (t), the carbon sequestration of other crops is 20.64×106 (t).The total CO2 storage of wetland is 609120t/a in Poyang Lake.There were some suggestions given, includes:For existing forest, tending wood scientifically and felling reasonably to form rational tree species structure, age structure and Lin layer structure, to improve the stability of forest ecosystem and carbon absorbing ability.Use the conservation tillage, water-saving irrigation and fertilization techniques, and to cultivate the new nitrogen efficient utilization of agricultural varieties, to improve photosynthesis and enhance carbon sequestration ability of local crops.Graze and ecological monitor the grassland resources rationally, control the size and number of breeding strictly, so as to achieve the goal of the grassland ecosystem in reducing and CO2 fixation.Prohibit silted up, perfect environment monitoring network, and also protect water wetland ecological functions.

    Crude extracts and fractions separated from ethanol extracts from Gelidium amansii and Sargassum pallidum were tested for their efficacy against bacterial by the circle agar disc diffusion method.As a result, Gelidium amansii and Sargassum pallidum had different degree of antimicrobial activitics.The antibacterial activities of crude extracts from Sargassum pallidum were stronger than antifungal activities.The ethanol extracts from Gelidium amansii showed the antifungal activities against Aspergillus niger with the inhibition zone 4 diameters, which were stronger than the antibacterial activities against Bacillus subtilis, Escherichia coli and Staphylococcus aurens.

    By investigating the contents of heavy metals as Cd, Pb, Hg, Cr, Cuand the harmful elements As heavy metal content in banana soil of Zhangzhou, Fujian province, evaluating them by taking the ways of single pollution index and comprehensive pollution index.It shows that, by taking the background value of soil of Fujian as the evaluation standard, soil of some of the banana lands of Zhangzhou are polluted by heavy metals with a pollution order of single pollution index as Hg (1.60) >Pb (1.26) >Cd (1.07) >Cu (0.76) >As (0.50) >Cr (0.38) and the main pollution elements as Cd, Pb and Hg, of which the pollution of Cd and Hg are moderate pollution;while the comprehensive pollution index is 1.16 that they rank light grade.By taking the second grade standard of soil environmental quality as the evaluation standard, the comprehensive pollution index is 0.34 that they rank non-pollution grade.Different sampling points on heavy metal pollution in a comparative analysis shows the road traffic and garbage and sewage on the banana plantations in the soil are caused by different levels of heavy metal pollution, mainly as Cd, Pb, Hg pollution.

    This experiment focused on testing the most suitable substrate for the tissue culture of Brassica alboglabra.The caulis segments and hypocotyl were separated from aseptic seedlings and used as explants.The results showed the best culture medium for hypocotyl was MS+6-BA 3.0 mg/L+NAA 0.2 mg/L, and for the caulis segments was MS+6-BA 5.0 mg/L+NAA 0.2 mg/L.The most suitable substrate for multiplication of adventitious buds was MS+6-BA 3.0 mg/L+NAA 0.2 mg/L, and the substrate with 1/2 MS+NAA 0.1 mg/L+IBA 0.2 mg/L had the best performance for rooting culture.The survival percentage of transplanting seedlings reached 96.6%.

    The effects of concentrations of KT and NAA on redifferentiation of Anemarrhena asphodeloides callus were studied using plant tissue culture methods and single-factor test.The results showed:The optimal rooting medium of A. asphodeloides callus was MS+KT 2 mg/L.The optimal regenerated shoot medium of A. asphodeloides callus was MS+KT 2 mg/L+NAA 1 mg/L.The optimal seedling growth medium of A. asphodeloides callus was MS+KT 2 mg/L+NAA 0.5 mg/L.The optimal proliferation medium of A. asphodeloides callus was MS+KT 2 mg/L+NAA 1 mg/L.

    Through the establishment of tissue culture rapid propagation system, the effects of KT and NAA on the induction of new stem, callus and roots of Astragalus membranceus were studied.The induction of new stem was related to the ratio of KT/NAA, which was obvious with the average number of new stem was the maximum, 11.7, when the value was 4.With the promotion of NAA, lots of white callus was produced.The average diameter of white callus, which was in the medium with 1 mg/L of NAA, was 1.4 cm.Roots came mainly from the white callus, being promoted by NAA but inhibited by KT.1 mg/L of NAA played a key positive role in the increase of roots number, the average length of roots and the number of secondary roots, but negative in roots average diameter.

    The current status of the vascular plants in Foshan City, Guangdong Province was reported in this paper.According to the inverstigation, a total of 1813 species belonging to 832 genera, 239 families of vascular plants are found in Foshan City.Among them, there are 96 species of Pteridophytas belonging to 63 genera in 40 families, 41 species of Gymnospermaes belonging to 28 genera in 13 families and 1676 species of Angiospermaes belonging to 979 genera in 278 families.There are also lots of wild plants in Foshan City, including 421 medicinal plants, 100 edible plants, 24 aromatic plants, 61 oil-bearing plants, 61 fiber plants, 8 starchiness plants, 1 resin plant, 30 tannic plants, 8 pigment plants, 90 timber plants and 123 ornamental plants.Among those, there are 15 national key protected plants and 20 rare or endangered plants classified as red list including Cibotium barometz, Alsophila spinulosa and so on.The suggestions of plant resources exploitation and protection were put out according to the natural and social conditions in Foshan City.

    Liliaceous and Orchidaceous plants are important plant resources and have positive significance for studying the system evolution and species diversity.In this paper, plant resources of Liliaceae and Orchidaceae from Mt.Maluanshan in Shenzhen, China have been investigated and studied, the resutes showed that Liliaceous plants has 14 species of 9 genera, which account for 64.3% and 70.0% of Liliaceous total number in Shenzhen;and Orchidaceous plants has 28 species of 22 genera, which account for 68.8% and 54.9% of Orchidaceous total number in Shenzhen.Based on the study of Liliaceae, Orchidaceae plants of Fenghuangshan Forest Park (3 genera and 3 species of Liliaceae;4 genara and 4 species of Orchidaceae), Tiegang Forest Park (3 genera and 3 species of Liliaceae;4 genara and 4 species of Orchidaceae), Tanglangshan Country Park (5 genera and 5 species of Liliaceae;8 genara and 9 species of Orchidaceae), Wutongshan Scenic Area (8 genera and 12 species of Liliaceae;17 genera and 30 species of Orchidaceae), Paiyashan Nature Reserve (7 genera and 8 species of Liliaceae;26 genara and 35 species of Orchidaceae), Qiniangshan Country Park (9 genera and 10 species of Liliaceae;21 genara and 30 species of Orchidaceae), whitch are on the central axis of the Shenzhen region from west to east, combined with the ecological environment feature, indicating the indicative significance in ecological environment and forest vegetation.Hereby authors put forward some ways for strengthening protection, management and research.

    Based on the field survey, the plant diversity of Changzhou Island, Guangzhou was studied.There are a total of 487 vascular plant species belonging to 358 genera, 127 families on the island, and the dominating vegetation are secondary evergreen broad-leaved forest and artificial vegetation.The diversity and geographical area of the wild plant on Changzhou Island were analyzed, the results indicated that few families are the dominant components of flora, and tropical elements predominate in the flora but there is a degree influence of temperate elements.Finally, suggestions for the protection of plant diversity were proposed.

    Yongchun County, locating in the transition between mid-subtropical and south subtropical areas, is rich in woody medicinal plant resources.Based on the field investigation, specimen collection and identification, and a large amount of references, the status of woody medicinal plants was studied.There are a total of 129 species (including varieties) woody medicinal plants in 90 genera of 45 families.The medicinal portions of woody medicinal plants and their corresponding functions were introduced briefly, and the suggestions about their protection and utilization were also put forward.

    According to the investigation on the vegetation in Mingdong District of Fujian Province, a newly recorded genus, Phalaris Linn., and four newly recorded species of Angiosperm including Phalaris arundinacea Linn., Saxifraga zhejiangensis Z. Wei. et Y. B. Chang, Brasenia schreberi J. P. Gmel., and Euphorbia maculate Linn. were found in Fujian.The voucher specimens are deposited in the Herbarium of Ningde Normal University (NDTC).

    To provide the reference for making quality standard of Fructus viticis drug, the quality of 16 Fructus viticis commodities from different drugstores in Fujian province were analyzed and evaluated.The source and trait of Fructus viticis commodities form different cities were collected and studied, and the contents of total ash, methanol-extract, total flavones, vitexicarpin were determined.The results showed that the quality of Fructus viticis commodities were influenced by manufacturing location, batch and the process of preparing Chinese medicine.It is necessary to make strict quality evaluation and control system at intermediate links to guarantee the effectual use of drug ingredients.

    Boron is a micronutrient essential for the normal growth and development of plants, and boron deficiency or toxicity is a worldwide agricultural problem, causing the decreases in yield and quality of crops.Since recent years, great progresses have been made in terms of molecular mechanisms of uptake and transport of boron in plants, which lays an important foundation for improvement of tolerances of plants to boron deficiency and toxicity by genetic engineering.In this paper, the progresses in molecular mechanisms and genetic regulations of uptake and transport of boron in plants were reviewed.
