Effects of Colletotrichum gloeosporioides Infection on Physiological and Biochemical Changes of Litchi Fruit
    LI Xin-yun, CHEN Wei-xin, LIU Ai-yuan
    2006, 35(01): 1-4.
    Abstract ( )   PDF (225KB) ( )
Changes in respiration rate, ethylene production, oxidation and peroxidation process and activities of several enzymes related to phenolic oxidation in litchi fruit were determined after inoculating litchi fruit with Colletotrichum gloeosporioides. The results showed that after inoculation, both respiration rate and ethylene production of litchi fruit enhanced significantly, the rate of superoxide radical anion (O2·) production and lipid peroxidation reactant-malondialdehyde (MDA) increased, the activity of superoxide dismutase (SOD) decreased while polyphenoloxidase (PPO), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) promoted on litchi pericarp. All above indicated that the infection of Colletotrichum gloeosporioides could enhance respiration rate and ethylene production, accelerate the course of oxidation and peroxidation, and induce the activities of POD, PPO and PAL on litchi pericarp. So the infection of Colletotrichum gloeosporioides was one of the important reasons that quickened senescence, browning and decay of litchi fruit.

Genetic Diversity Analysis on Eight Turfgrass of Poaceae by RAPD
CHEN Yan, HU Mu-lin, YUAN Chang-chun, GUAN Guang-hui, LUO Mei, HUANG Yong-lian
    2006, 35(01): 5-8,19.
    Abstract ( )   PDF (236KB) ( )
The genetic diversity of eight turfgrass varieties representing four species in four genera of family Poaceae was analyzed using random amplified polymorphic DNA (RAPD) method.Fifteen 10-meroligonucletide primers were used for DNA ampification, and 144 DNA bands were generated, of which 63.2% is polymorphic loci. The genetic distances are 0.0857~0.2928 among eight turfgrass varieties, the maximum value is between the variety Crossfire Ⅱ of Festuca arundinacea and Accent of Lolium perenne, the minimum value is between the two varieties of Unhulled Cynodon dactylon and hulled Cynodon dactylon. The two dendrograms based on genetic distance by UPGMA and Neighbor-joining method are similar topologically. The relationships among different varieties from the same species are very close. The relationships of inter-genera or interspecies are relatively far, although the different genera of the tribe Chlorideae get together into a clade. The four warm-season turfgrass form a clade. The variety Accent of Lolium perenne, a cool-season turfgrass, has a far relationship to other turfgrass and forms a clade. The study didn't support to put the genus Lolium and Festuca into the same tribe Poeae and suggested that the turfgrass Festuca arundinacea be treated as an intergrade type of turfgrass.

Influence of Medium and Light Intensity on the Leaf Explant Browning in Phalaenopsis sp.in vitro
    XU Chuan-jun, LI Ling
    2006, 35(01): 9-12.
    Abstract ( )   PDF (216KB) ( )
The browning ratio of Phalaenopsis sp.leaf explant in vitro growing on MS liquid paper bridge and 1/2 MS after 10 d is 0 and 67%, respectively, less than that growing on MS, B5 and N6.Doubling the content of FeSO4 or microelement deficiency could increase the browning rate of explant cultured in MS medium.Reducing the content of agar could increase the degree of browning. The higher the intensity of light, the more serious the explant browning.Explant cultured on medium supplemented with citric acid showed a considerable reduction in browning.Adding Vit C, active carbon or PVP could not overcome the browning of explant.

    2006, 35(01): 25-28.
    Abstract ( )   PDF (1644KB) ( )
The anther of Malvavisus arboreus var.penduliflorus consists of epidermis (one layer), endothecium (one layer), middle layer (two layer), tapetum (one layer) and aporgenous cells. The anther has four chambers and the development of anther wall is dicotyledonous type. The microsporogenesis and male gametophyte development are observed as follows:with the meiosis of microspore mother cells, tetrad is formed, of whish the cytokinesis is simultaneous type and the arrangements are tetrahedral, crossing and left-right symmetrical;the microspores complete the mitosis and generate double-nucleus pollen;the generative nucleous complete the mitosis again and generate three-nucleus pollen. The eoidermis disappears at the uninuclear microspore stage, the middle layer disappear at the pollen mother cell stage, the endothecium forms fibrous layer at uninuclear microspore stage, and the tapetum cells disappear at the uninuclear microspore stage and the tapetum is amoeboid one.The female gametophyte development is observed to indicate epigynous ovary, five locules, one ovule per locule, carpylotropous, axial placenta;most of ovules development stop at the nucellus formation stage and a few of nucellus form many archesporied cells, uninuclear and double-nucleus embryos.
