    HUANG Cong-li, LI Chuan-yong, PAN Ai-min, XU Wei-dong, XU Rui-de
    2001, 30(03): 1-6.
    Abstract ( )   PDF (931KB) ( )
Genetic DNA diversity among thirteen cauliflower cultivars were analyzed using Random Amplified Polymorphic DNA (RAPD). There were 175 clear and reproducible bands amplified with twenty primers and 118 of these bands were polymarphic(67.4%). The genetic similarity coefficients between different cauliflower cultivars based on RAPD markers were analyzed and the dendrogram of different cultivars based on UPGMA analysis was also generated. The results showed that the genetic diversity among different cultivars was abundant. The genetic relationship among cultivars, application of RAPD technique in differentiation and breeding of cauliflower were preliminary discussed.

Protective effect of ASA on membrane damage in protoplast of peanut during enzymatic isolation of leaf protoplasts
    FU Ming-hui, ZHUANG Dong-hong
    2001, 30(03): 7-10.
    Abstract ( )   PDF (166KB) ( )
Malondialdehyde(MDA) accumulated in protoplast of mesophyll cell of Arachis hypagaea during the cell wall digestion in enzyme solution. Added with ascorbic acid (ASA), the content of MDA decreased. At the initial stage of enzyme digestion, the activities of peroxidase (POD) and catalase (CAT) increased without adding ASA and after a long time of enzyme digestion, the activity of POD decreased. While ASA added into enzyme solution, the activity of superoxide disumutase (SOD) increased significantly. These results showed that membrane were damaged during the protoplasts isolation; cells had a protection mechanism to respond to it and adding ASA could reduce the membrane damage during protoplasts isolation.

    Abstract ( )   PDF (175KB) ( )
Effects of 2h, 5d, and 30d of storage at 5℃ on the shelf quality and physiological aspects of Huaizhi litchi were investigated. The results showed that Huaizhi litchi kept at 5℃ cool storage resulted in a significant decrease of fruit quality and an increase in both relative permeability of pericarp cell membrane and peroxidase activity when compared to the control. The change of carbon dioxide and ethylene evolution of litchi in sealed container during shelf life was also discussed.

    Abstract ( )   PDF (220KB) ( )
The effects of 5 kinds of food additive and 7 kinds of metallic ion on the stability of red pigment from Rubus lambertianus were investigated. The results showed that glucose, sucrose and sodium benzoate had no adverse effects on the stability of red pigment and glucose and sucrose showed a certain fortification; citric acid could increased the stability significantly; Vitamin C could promote the oxidation of pigment. Na+, Mg2+, Al3+, Zn2+ had no effects on the stability of R. lambertianus red pigment while Mn2+ showed definite adverse effect on it and Cu2+, Fe3+ decreased the stability significantly.

    2001, 30(03): 26-27,49.
    Abstract ( )   PDF (134KB) ( )
The experiments with different grafting methods, taking the annual seeding of wild kiwifruit (Actinidia deliciosa) as stock and new cultivar's (Jinkui) one-year shoot as scion wood, were conducted. The results showed that the survival percent of whip grafting and side grafting with single bud scion reached 98.4% and 96.1%, respectively, and the growth of seedlings, i.e. basal diameter, new shoot length, leaf number, was significantly better than those with other grafting methods. They are convenient to operate and can be popularized into forestry production.

    2001, 30(03): 28-32.
    Abstract ( )   PDF (196KB) ( )
There are 36 rare and endangered species in Nanling National Nature Reserve, Guangdong Province, making up 9.3% of the total in China (388 species), including 8 gymnosperms and 28 angiosperms. Among them, 2 are endangered, 20 vulnerable and 14 rare species. The resources of rare and endangered plants in Nanling National Nature Reserve possess the features of richness in composition, obvious tropical nature, antiquity of origin, abundance of endemic elements and diversity of usage. Based on the analysis of characteristics of the resources, some protective recommendations are put forward.

    Abstract ( )   PDF (132KB) ( )
The effects of different concentrations of various hormones on the root number and length of Limonium bicolor were studied and the survival ratio and growth of plantlets in different substrates were evaluated. The results showed that MS+NAA 1.0mg/L gave the best result in root number, MS+IBA 1.0mg/L was best for root length, and the plantlets transplanted in peat and perlite substrate (1:1) resulted in the best growth. The cost of rooting and transplanting of plantlets of Limonium bicolor was also discussed.
