银叶树品种‘Kelvin Red’启动培养研究

摘要: 以银叶树‘Kelvin Red’为试材,对其离体快繁启动培养过程进行了研究。试验结果表明:银叶树外植体最佳的消毒方案为75%酒精浸泡30s,再用0.1%升汞进行5+4 min的二次消毒;最佳取材季节为冬季;最佳外植体类型为茎尖和茎段中上部半木质化部分;最佳培养基配方为WPM+6-BA 1.0 mg/L + IBA 0.5 mg/L。

Abstract: The initiation culture of in vitro rapid propagation in Leucadendron ‘Kelvin Red’ was investigated. The results showed that the best method for explants disinfection was as follows: the explants were surface-disinfested in a solution of 75% alcohol for 30 seconds and then dipped into 0.1% mercuric chloride two times for 5 minutes and 4 minutes respectively. Winter was the optimal season for collecting explants. It was found that shoot tips and the upper semi-lignified stems were the optimal explants materials. The optimal startup medium for in vitro Leucadendron propagation was WPM + 6-BA 1.0 mg/L + IBA 0.5 mg/L.
