罗汉果伯林2号种子形态结构及萌发特性

Seed Morphology and Germination Characteristics of Siraitia grosvenorii Bolin No. 2

摘要: 以广西桂林永福罗汉果Siraitia grosvenorii伯林2号种子为材料,测定种子形态、千粒重、吸水率、发芽率和发芽势,以及不同浸种处理、pH、PEG-6000浓度对其萌发的影响。结果表明,罗汉果伯林2号种子呈扁平状类圆形,长17.00 mm,宽14.34 mm,厚3.44 mm,千粒重178.71 g。罗汉果种子吸收的水分主要储存在种壳边缘厚壁纤维的缝隙和死亡的海绵细胞中。其成熟种子在25 ℃、相对湿度60%、12 h光照/12 h黑暗条件下发芽率为46.67%,发芽势13.33%。浸种可使种子发芽始末时间提前7~8 d。罗汉果种子萌发显著受酸胁迫和干旱胁迫影响。当在弱酸性(pH 6)条件下,其发芽率最高53.33%,当pH降低,其发芽率和发芽势均显著降低。高浓度PEG-6000模拟干旱胁迫不仅降低种子发芽率和发芽势,还显著影响幼苗长势。

Abstract: The seeds of Siraitia grosvenorii Bolin No. 2 from Yongfu, Guilin, Guangxi were used as experimental materials to study the germination characteristics. The morphology, thousand-grain weight, water absorption rate and germination rate of the seeds were observed and measured, as well as the effects of different soaking treatment, pH, and PEG-6000 concentrations on the germination of S. grosvenorii seeds. The results showed that the seeds of Bolin No. 2 were flat-shaped and round-like, with a length of 17.00 mm, width of 14.34 mm, and thickness of 3.44 mm, and the thousand-grain weight of 178.71 g. The water absorbed by the S. grosvenorii seeds was mainly stored in the large gaps in the thick-walled fibers at the edge of the seed coat, which was needed for seed germination and growth. The mature seeds had a germination rate of 46.67% and a germination potential of 13.33% under the conditions of 25 ℃, 60% relative humidity, 12 h light/12 h dark. Soaking could advance the start and end time of seed germination by 7–8 days. The germination of S. grosvenorii seeds was significantly affected by acid stress and drought stress. Its germination rate was highest (53.33%) under weakly acidic conditions (pH 6), while acidity continued to increase, its germination rate and germination potential were significantly reduced. High concentration PEG-6000 drought stress not only reduced the germination rate and potential, but also significantly affected the growth vigor of seedlings.
