    Inhibition Effects of Four Indole Alkaloids in Voacanga africana on Tumor Cells in vitro

    To investigate the anti-tumor activities of four indole alkaloids, Vincamine, Coronaridine, Voacamine and Voacangine in Voacanga africana in vitro, the proliferation inhibition effects of SKOV3, BEL7402, SMMC7721 and Changliver cells were determined by MTT assay, and the morphological changes of apoptosis were observed by AO/EB staining. The results of MTT assay showed that there was a dose dependent relationship between the proliferation inhibition of four cell lines and the four indole alkaloids. The growth inhibition rates of 50 μg·mL-1 Voacamine on all four cell lines were all over 95%; at the same concentration, Voacangine only inhibited BEL7402 and Changliver by more than 78%; Coronaridine had a proliferation inhibition rate of only 50% for Changliver; the effect of Vincamine on the proliferation of four cell lines was not obvious. After staining by AO/EB method, the four cell lines showed nuclear shrinkage, concentration and migration under the action of 12.5 μg·mL-1 Voacamine, indicating that Voacamine had obvious effect of inducing apoptosis; Voacangine only had the same effect on BEL7402 and Changliver, and the other two alkaloids acting cell lines showed no obvious apoptosis. The MTT and the AO/EB staining method showed consistent results. Voacamine and Voacengine could induce apoptosis of ovarian cancer cells and human hepatocytes, and anti-tumor effects of the other two alkaloids were relatively weak.

    Analysis of Volatile Components of Bombax ceiba Flower by HS-SPME-GC-MS

    The volatile components of Bombax ceiba flower were analyzed by HS-SPME-GC-MS method. Fifty-six volatile organic compounds were identified by GC-MS and their relative contents were also calculated by area normalization method. It was found that the main volatile components of B. ceiba flower were Modephene (13.74%), (1R,3aS,5aS,8aR)-1,3a,4,5a-tetramethyl-1,2,3,3a,5a,6,7, 8-octahydrocyclopenta[c]pentalene (10.95%) and 2-methylene-4,8,8-trimethyl-4-vinyl-bicyclo[5.2.0] nonane (10.36%). Among these components, terpene and alkyl compounds were accounted for 55.24%, 29.72%, respectively, as well as small quantities of aldehyde, ester and ketone constituents. The results of this study could provide an experimental basis for development and utilization of B. ceiba flower.

    In order to carry out the isolation and identification of the differential proteins under the cold and drought stress of tea plants, using the strong stress-resistant tea tree variety ‘Yingshuang’ as experimental material. The two-dimensional gel electrophoresis technology system suitable for leaf was screened by extraction method, IPG strip pH range, protein content, separation gel concentration and staining method. The results showed that total protein was extracted by TCA-acetone or Tris-HCl method, using 17 cm pH 4—7 IPG strip and 1.6—2.2 mg protein for isoelectric focusing, using 13.5% polyacrylamide gel for electrophoresis, subsequently staining by high sensitivity Coomassie Brilliant Blue R-250 method. Finally, the protein of each molecular weight of the leaf was sufficiently separated, and a 2-DE pattern with high resolution, clear background and good reproducibility was obtained. The system was suitable for the analysis of differential proteins in leaves of ‘Yingshuang’ under cold and drought stress.

    Biological Characteristics and Active Ingredients Contents of Anoectochilus roxburghii from Guangxi at Different Months after Transplantation

    This study investigated the biological characteristics and changes of polysaccharide, polyphenols and flavonoids of Anoectochilus roxburghii from Guangxi at four-month to nine-month plants after transplantation. Results were as follows: there was a certain increase in the biomass of A. roxburghii as the increase of transplanted month, and the biomass of nine-month after transplantation was highest. The change trend of polysaccharide content was first decreased and then increased, which was similar to the ‘V’ shape. There was no significant difference between four-month and nine-month after transplantation, and the highest polysaccharide content appeared at 4 months after transplantation. The highest contents of polyphenols and total flavonoids were appeared at 6 months after transplantation.

    In this study, the content of photosynthetic pigments and chlorophyll fluorescence kinetics parameters of normal green leaves and yellow mutation leaves of Cymbidium ensifolium ‘Ba Bao Qi Zhen’ were compared. Results showed that the content of chlorophyll (Chl), chlorophyll a (Chl a) and chlorophyll b (Chl b) of yellow mutation leaves were significantly lower than those of the normal green leaves, and the contents showing a gradual decline with the increase of mutant area. Furthermore, it was found that the initial fluorescence (Fo), maximum fluorescence yield (Fm), Kautsky effect maximum fluorescence (Fp), the steady state photochemical quenching coefficient (qP) and light adaptation steady-state fluorescence yield (Ft_Lss) of yellow mutation leaves were significantly lower than those of the normal green leaves, while there was no significant difference in PSⅡ original light energy conversion efficiency (Fv/Fm). In addition, the steady state of photochemical fluorescence quenching coefficient (NPQ) of yellow mutation leaves was higher than that of the normal green leaves.

    Diurnal Changes of Photosynthesis and Fluorescence Parameters in Camellia amplexicaulis under Different Light Intensities

    In order to study the effect of different light intensities on the photosynthesis mechanism of Camellia amplexicaulis, we detected the diurnal changes of chlorophyll fluorescence parameters of shade in 3 months respectively. The results showed that under different light intensity treatments, the net photosynthetic rate presented a single-peak curve with the peak at around 11:00am. The decrease of photosynthetic rate was mainly caused by stomatal limitation factor. The chlorophyll fluorescence parameters indicated that electron transfer was blocked, the heat dissipation increased, the PSII reaction center was destroyed under strong light conditions, which reduce the photosynthetic efficiency of C. amplexicaulis; under excessive shade condition, photoinhibition of C. amplexicaulis resulted in damage of photosynthetic apparatus; the photosynthetic efficiency was higher under 45% illumination. Correlation analysis showed that Pn was significantly correlated with Ta, RH, Gs, Tr, WUE and ETR, and was significantly negatively correlated with RH and negatively correlated with Ci. The main environmental factors affecting the net photosynthetic rate of C. amplexicaulis were the temperature and relative humidity of air.

    Seed Germination of Four Species of Wild Primula

    Taking four species of wild Primula (P. polyneura, P. sikkimensis, P. bulleyana and P. pulverulenta) as the experimental materials, the influence of the seed morphology, light, and GA3 on their seed germination was studied. The results revealed that the seed morphologies of four species of Primula were various. The thousand seed weight of P. sikkimensis?was the largest (0.458 g); while that of P. pulverulenta?was the smallest (0.192 g). Light and GA3 treatments with different concentrations had different influences on the seed germination of four kinds of Primula. The effect of light on seed germination of P. polyneura?was the best, followed by P. sikkimensis?and P. bulleyana. However, it had no obvious promoting effect on P. pulverulenta. Under the condition of light, the treatment with 400 mg·L-1 GA3 had a better germination effect on P. bulleyana?and P. pulverulenta, which significantly increased the germination rate, and shortened the germination duration. The suitable GA3 concentration for P. sikkimensis?seed germination was 200 mg·L-1.

    Species composition and diversity of the broad-leaved forest and shrub communities with Camellia japonica were studied based on the investigation of typical plots. The results showed that a total of 147 species of vascular plants, belonging to 118 genera in 65 families, were recorded in 8 plant communities. The geographical elements were various, while the tropical elements outweighed the temperate and subtropical elements,?which showed the transitional characteristics from subtropical to warm temperate and reflected the relative advantage of habitat thermal condition. In communities with Camellia japonica, the species richness and Shannon-Wiener index of the arbor layer presented the significant negative correlation with the succession trend. The species richness and Shannon-Wiener index of shrub layer and liana layer in the higher altitude of shrub and sparse forests were greater than those in the broad-leaved forest. Effected by prominent dominant of the dominant species in corresponding communities, including dominant species Machilus thunbergii, Cyclobalanopsis glauca and Distylium gracile in arbor layer, or dominant species Neolitsea sericea in regeneration layer, the variance of Pielou index of arbor layer and regeneration layer was great.

    Floristic Characteristics of Lycophyte and Fern Flora in Jingning, Zhejiang Province

    Based on field investigation, the paper preliminarily discussed the floristic characteristics, geographical distribution, ecological environment, relationships with adjacent regions and species endangerment assessment of lycophytes and ferns in Jingning She Autonomous County. 296 species, seven varieties and one hybrid species belonging to 83 genera and 29 families were discovered, including nine new distribution records in Zhejiang Province. The geographical elements of lycophytes and ferns were mainly of tropical nature as to the genera, and the species were mainly of temperate nature. This area belonged to the convergence zone of subtropical and temperate species. Compared with the adjacent provinces and regions, the locophyte and fern flora of Jingning were most similar to those in Jiangxi Province, Fujian Province, Ningbo City and Wenzhou City. They were more close to the flora of Wenzhou and less with Anhui. The ecological types of lycophytes and ferns in Jingning could be roughly divided into six categories: warm coniferous forest habitat, broad-leaved evergreen-forest shady and wet habitat, bamboo thicket warm and wet habitat, temperate pine, fir and cypress cold and wet habitat, drought habitats near shrubs and village roadsides, wetland habitats in streams, farmlands and marshes. Among them, the broad-leaved evergreen-forest shady and wet habitat had the highest diversity of species. There were 46 threatened lycophytes and ferns in Jingning, Zhejiang Province, including 11 endangered species (EN), 35 vulnerable species (VU) and four of them were provincial protected species.

    Ornamental Characteristics Evaluation of Cymbidium sinense Cultivars Using the Principal Component Analysis

    By measuring and observing 20 ornamental traits including plant height, flower color and petal form etc. of 30 cultivars of Cymbidium sinense cultivated in Fuzhou for several years, the variation range, correlation and principal components of 20 ornamental traits of C. sinense were analyzed. Ornamental characteristics of 30 varieties of C. sinense were also analyzed by applying principal component analysis. The results showed that 16 quantitative traits had varying degrees among different cultivars, and there were large differences in flower morphology, but small variation in leaf morphology and quantity. Besides, the genetic diversities of flower color and leaf were high. There was a significant correlation between leaf morphology and flower morphology. 20 ornamental traits could be divided into 7 principal component factors, named plant height factor, petal form factor, flower color factor, leaf number factor, flower diameter factor, leaf width factor and leaf art factor. The contribution rate accumulated of the variance of 7 principal component factors reached to 87.35% of the total trait information. By calculating and sorting the important principal component values of each variety, four varieties with excellent ornamental traits were ‘Baimo’, ‘Baoshanzhua’, ‘Longmei’ and ‘Jinhuashan’.

    The ornamental traits of 35 Dendrobium species, including flower length, flower color, floral, flowering amount, flower morphology, leaf morphology and plant morphology, were surveyed and recorded in this paper. 35 Dendrobium species were classified according to the flower length, aroma concentration, flowering amount and flower size, then comprehensive evaluation of six species of flower-viewing varieties with better performance in all aspects.

    Based on the introduction and cultivation performance of 10 species of Labiatae aromatic plants in Fuzhou, 15 evaluation factors were screened from three aspects: ornamental value, development potential and stress resistance, and a quantitative comprehensive evaluation was made, and its adaptability and landscape application in Fuzhou were also evaluated. The results showed that the comfort of smell, processing value, leaf color, flower color, repellency and heat tolerance of aromatic odor were the main indexes for comprehensive evaluation of aromatic plants in Fuzhou. The aromatic plants with the highest comprehensive score were Thymus mongolicus, Rosmarinus officinalis and Mentha spicata, which had broad application prospects in landscape.

    As the main place for public to carry out outdoor recreation activities, urban garden green space is an important carrier for shaping high-quality environmental education. This article analyzed the public cognition of the function of green space in Guangzhou in the form of online questionnaires and on-site questionnaires and interviews, in order to provide reference for the construction of environmental education sites. The results showed that the public cognition of specific descriptions of species diversity and landscape diversity tended to be consistent. The large majority of the public believed that green space vegetation could play an important ecological role. And the public education was higher, their desire to recognize the garden plants became stronger. Most people chose the semi-open spaces with plants densely populated in as resting place. In the design of garden green space, by enhancing the convenience and interest of popular science information, it would work better in the environmental education of green space.

    Evaluation of Campus Plant Landscape by UAV Aerial Photography——Taking South China Agricultural University as an Example

    Based on unmanned aerial vehicle (UAV) aerial photography, plant application status investigation and questionnaire survey, eight kinds of plant landscape types of South China Agricultural University were evaluated using beauty degree evaluation method. Results showed that plant community stratified structure in South China Agricultural University was clearly visible, space layout was abundant, overall level of plant landscape was good. Plant color richness and plant diversity were significantly correlated with plant landscape, which were effective factors affecting the landscaping value of campus plants. The research results provided a reference for the evaluation and construction of plant landscape.

    During the fourth survey of Chinese medicine resources in Wuyishan City and Pucheng County of Fujian Province, six new records of plants in Fujian province were discovered, Vicia kulingana, Aster jiulongshanensis, Epimedium leptorrhizum, Scutellaria tenera, Neanotis hirsuta, Rubus parvifolius var. toapiensis, among which Rubus parvifolius var. toapiensis was a new recorded variety in Chinese mainland. The discovery of six new recorded plants in Fujian further enriched the ecological diversity of Fujian province and provided new information for the study of Fujian flora.

    Growth Effect of Different Sizes of Root-controlled Containers on Rubber Mini-seedling Budding

    The growth vigor of rubber mini-seedling budding was observed by suspension and near-ground culture with different specifications of root-controlled containers. The results showed that suspension culture had a significant effect on the longitudinal elongation (such as plant height and main root length) of rubber grafted seedlings. When the height of seedling containers was the same, the larger volume of seedling containers, the more extension space of root system and better growth of the seedling’s above-ground part. When the volume was the same, the higher height of seedling containers, the longer length of seedling’s main roots. But there were little influence on the above-ground part of seedlings and the diameter of main roots.

    Using the plant without diseases and insect pests as experimental material, the renewable technologies of stem section, leaf in Tripterygium wilfordii were studied by orthogonal experimental design. The results showed that the optimal medium for bud induction was MS + 6-BA 1.5 mg·L-1 + IBA 0.1 mg·L-1; the optimal medium for subculture was MS + 6-BA 1.0 mg·L-1 + NAA 0.1 mg·L-1; the best culture medium for seedling cultivation was MS + 6-BA 0.5 mg·L-1 + VB1 0.5 mg·L-1 + Peptone 0.5 g·L-1; the optimal rooting medium was 1/2MS + NAA 2.0 mg·L-1 + KT 0.1 mg·L-1. In transplanting, the best substrate ratio was vermiculite and peat mixed with 1:1, and 30 days after transplanted, the transplanting survival rate was up to 84.5%.
