    Expression and Antibody Preparation of Sugarcane Mosaic Virus P1 Protein

    The target gene was subcloned into the prokaryotic expression vector pET-32a,and transformed into E. coli Bl21(DE3),to obtain recombinant pET-32a-P1.Ultrasound results showed that the fusion protein existed in soluble form.Expression product had the same size with the target protein detected by SDS-PAGE.The antibody against the P1 protein was gained by immunized rabbit with target protein cut from SDS-PAGE.Indirect ELISA measurements proved that the antibody specificity was fine,because of that the dilution of 51200-fold not merely kept positive still,but also passed western blot.

    Bougainvillea glabra ‘Formosa’ is an ornamental which has healthy function.To explore the mechanisms of the secondary-metabolism in Bougainvillea,the volatile components in leaves of Bougainvillea glabra ‘Formosa’ was extracted by steam distillation and were analyzed by GC-MS.In this study,61 chemicals were isolated.After searching NIST database,47 chemical components were identified,which consists of 90 percent of naphtha.The results showed that Bougainvillea glabra ‘Formosa’ had much health-friendly chemical including 2-Hexenal,Phytol,n-Hexadecanoic acid,2-Methoxy-4-vinylphenol,D-Limonene and so on.

    Microscopic observation on two ‘soft core’ hawthorn germplasms (C. pinnatifida ‘Xinbinruanzi’ and C. pinnatifida ‘Kaiyuanruanzi’) and two ‘hard core’ hawthorn germplasms (C. pinnatifida ‘Chuizhishanlihong’ and C. pinnatifida ‘Duanzhishanlihong’) were carried out in their rigid period.The results showed that the accumulation status of lignin in starting rigid period could be identified rapidly by Wiesner method.Through this experiment,some fundamental data was given for differential gene expression and formation mechanism of hawthorn ‘soft core’.

    The investigation and analysis were performed to study the flowering characteristics of ‘Caomeili’ from single ear,single plant and the group of ‘caomeili’ plants.The results showed that the flowering order was female flower to male flower.In other words,the female flower opened in the first period and the male flower second;but the flowering order of few flowers was male flower to female flower to male flower.Both the flowering period and the time of male flower meeting with female flower of the group were longer than those of single ear and single plant,which helped pollination and impregnation.The first peak of fruit drop came after flowering for 18-25 d,when the amount of fruit drop was the largest;and the second peak came after flowering for 51-58 d.

    Effects of Salt-Stress on the Growth and some Physiological and Biochemical Indices of Dioscorea bulbifera Virus-Free Plantlets

    Dioscorea bulbifera virus-free plantlets were treated with different concentrations of NaCl to study the effects of salt stress on their growth and related physiological and biochemical indices.The results showed that the growth of Dioscorea bulbifera virus-free plantlets was markedly retarded under salt-stress.With the increasing of concentration,the total chlorophyll content and the activity of SOD decreased,while the contents of malonaldehyde (MAD) and proline increased.Moreover,under the salt-stress conditions,the content of proline in leaves of Dioscorea bulbifera virus-free plantlets had a significantly positive correlation to the content of MDA,but a significantly negative correlation to the total chlorophyll content and the activity of SOD.It was concluded that the changes of proline content in leaves could be used as a physiological index to reflect the hurt degree of Dioscorea bulbifera virus-free plantlets under salt-stress.

    Influence of 5 Plant Growth Inhibitors on in vitro Conservation of Endangered Emmenopterys henryi Germplasm

    The stem segments with buds of Emmenopterys henryi were used as explants for studying the effects of 5 plant growth inhibitors at different temperature and light conditions on the in vitro conservation by single-factor experimental method.The results showed that at 25℃,the best CCC,MH,B9,S3307 and ABA concentrations of Emmenopterys henryi germplasm in vitro conservation were 4 mg/L,4 mg/L,2 mg/L,1 mg/L and 4 mg/L.At 9℃,the best CCC,MH,B9,S3307 and ABA concentrations were 2 mg/L,1 mg/L,1 mg/L,0.5 mg/L,and 2 mg/L.On light or dark conditions,the best CCC,MH,B9,S3307 and ABA concentrations were 4 mg/L,4 mg/L,2 mg/L,1 mg/L,and 4 mg/L.Compared with normal temperature conditions,5 plant growth inhibitors had higher survival rate and their best concentrations for Emmenopterys henryi germplasm in vitro conservation generally reduced on low-temperature conditions.And compared with light conditions,5 plant growth inhibitors also had higher survival rate on dark conditions,but their best concentrations were the same as light conditions.The genetic variation of Emmenopterys henryi in vitro conservation plantlets did not appear.

    ISSR-PCR markers were used to analyze the genetic diversity of 40 wampee cultivars.A total of 15 ISSR primers selected from 96 primers were used to amplify 165 DNA fragments and of which 100(60.6%) were polymorphic loci.Based on SPSS 13.0 analysis,the similarity coefficient between different cultivars ranged from 0.714 to 1.000.The dendrogram constructed by UPGMA showed that 40 wampee cultivars were classified into five groups.

    The orthogonal design was used to optimize SRAP-PCR amplification system on Clausena lansium on four levels of five factors (Taq polymerase,Mg2+,DNA template,dNTPs,and primer,respectively).The results indicated that the order of each factor in different levels affected on the result of PCR was Mg2+ or Taq polymerase,DNA template,primer and dNTPs.An optimal SRAP-PCR system was established,that was total 25 μl reaction system containing PCR10×Buffer 2.5 μl,0.75U DNA polymerase,2.0 mmol/L Mg2+,60 ng DNA template,0.2 mmol/L dNTPs,and 0.2 μmol/L primer.

    The effects of different shading treatments (0,50%,75% and 90% of full light) on the growth and photosynthetic characteristics of Clerodendranthus spicatus were studied.The results indicated that compared with full light,the net photosynthetic rate and transpiration rate of leaves treated by three kinds of shading treatments decreased,leaves thickness and their palisade tissue were thinner,fresh weight and branch number reduced,sections spacing and plant height increased significantly,and increment of roots,stems and leaves declined markedly.In sum,the most suitable light condition for the growth of Clerodendranthus spicatus was full light.

    The Sensitivity of Zoysia sinsca on Kanamycin

    The effects of kanamycin with different concentrations and different treatment times on seed germination and seedlings growth of Chinese Zoysia were studied by indoor germination experiment.The results indicated that the difference in seed germination between treatments with kanamycin and without kanamycin was not significant,and the differences of different concentrations and different treatment times on the growth of seedlings were significant.With the increasing of kanamycin concentration,the plant height,root growth and lateral root growth of Zoysia were inhibited,and albinism rate increased at the same time;With the prolonging of treatment time,plant height and root growth decreased.According to comprehensive index,75 mg/L kanamycin treated for 6~10 d gave the best result for the growth of Zoysia sinica seedlings.

    Effects of Postharvest Temperature on Defoliation and Stress-Resistant Physiology of Potted Ficus ginseng

    Potted Ficus ginseng was used to study the effects of different postharvest temperature on the changes of defoliation and stress-resistant physiology during storage and transportation.The results demonstrated that the best postharvest temperature could significantly reduce membrane permeability and malondialdehyde (MDA) content of leaves,postpone the degradation course of penetrative adjustive substance in leaf and the descent of chlorophyll content and poly-phenols aegis enzyme activity of leaves,improve anti-stress capability during storage and transportation,delay Ficus ginseng decrepitude,and reduce the defoliation rate (<16% defoliation rate after 36 days' storage and transportation at 10±1℃ and 13±1℃).Moreover,lowering the temperature gradually,2℃ each day,to storage and transportation's temperature before storage and transportation at 10℃ could reduce defoliation rate after 36 days' storage and transportation.

    The method and influencing factors of total flavonoids extraction in Juglandis Mandshuricae Cortex were studied systematically.Taking the total flavonoids content as index,the effect of extraction method including routine extraction,ultrasonic extraction,reflux extraction and Soxhlet extraction and kind of solvent,concentration and temperature on extraction rate were investigated.The results showed that the best extraction method was ultrasonic extraction.The extraction rate was the highest when the concentration of ethanol was 60% and the temperature was 60℃.

    The investigation of characters of Manglietia patungensis Community in Guanshan National Nature Reserve of Jiangxi Province showed that:there are abundant species components and complicated floristic elements in M. patungensis community.In terms of genus geographical composition,more of them are distributed in tropical zones.According to Raunkiaer's statistics of life-form,the phaenerophyte in the community composition take primary position and the hemicryptophyte are the second.Regarded to the leaf characteristics,the community is mainly composed of mesophyll,microphyll,simple leaf,sclerophyll and unentire leaf.The vertical structure of community is relatively complicated,where the species diversity of undergrowth layer is apparently greater than tree layer.The structure of whole M. patungensis community is stable and contagious distribution.

    Based on the field investigation,there are 37 Chinese endemic genera (including 3 endemic genera to East China),40 species of seed plants belonging to 30 families in Sanqingshan Mountain,which occupies 13.75% of the genera in China,53.62% of the genera in eastern China and 66.07% of the genera in Jiangxi Province.It is one of the most conspicuous areas for the biodiversity of Chinese endemic genera of seed plants in Jiangxi Province and eastern China even.The dominant flora of Chinese endemic genera in Sanqingshan Mountain is dicotyledon with mainly life forms of deciduous trees and shrubs and perennial herbages.For geographic distribution,Chinese endemic genera in Sanqingshan Mountain keep close relationship with the adjacent Mountains of east China.The traits of their origin of species are isolated and original,and there are also many endangered species inside.

    Investigation was carried out on ornamental pteridophyte resources in Meihua Mountain Nature Reserve,which located in the west of Fujian Province.There are 68 species of wild ornamental pteridophyte plants,belonging to 29 families,48 genera.According to their application,they are classified into 4 categories including landscaping,potted landscape,cut-flower and pot culture.Moreover,their species composition,horizontal,vertical and ecological distribution,superior resources and utilization ways are also analyzed in this study.Some suggestions are put forward for the development and utilizationof the ornamental pteridophyte as well.

    The trial used the original seedling stems of dharma orchid for tissue culture technology study,selected out the optimum medium:(1) protocorm induced:MS+6-BA 0.5 mg/L+NAA 1.0 mg/L+banana 250 g/L;(2) bud induced:1/2 MS+6-BA 3 mg/L+NAA 2.0 mg/L+AC 2.5 mg/L+su 3%+banana 200 g/L;(3) shoot proliferation:1/2 MS+6-BA 3 mg/L+NAA 2.0 mg/L+AC 2.5 mg/L+su 3%+banana 150 g/L;(4) rooting:1/2 MS+6-BA 0.3 mg/L+IBA 0.5 mg/L+NAA 0.5 mg/L+AC 2.5 mg/L+su 3%.Using mixture of perlite,fine stone and bark as culture substratum,the survival rate was up to 95%.

    The disinfection method of seed and culture condition of aseptic seedling in tissue culture of Tetracentron sinensis were optimized in this experiment.There were 4 disinfection methods used in total and the results were as follows:the germination of T. sinensis seed was inhabited strongly by 0.1% HgCl2 and weakly by 4% NaClO;the best disinfection method was 4% NaClO.The culture medium with moderate salt and low nitrogen was in favor of the germination of seed,and 1/4 MS medium was the optimal medium for culturing aseptic seedling of T. sinensis.

    Effect of the Construction of Green Belts on the Attenuation of Traffic Noise along the Urban Trunk Roads in Xiamen City

    Through site investigation and noise monitoring,the effects of the construction of green belts along the urban trunk roads in Xiamen Island on the attenuation of noise has been studied.The results show that the construction of green belts along the trunk road can be divided into four kinds of types as bellows:type I,single arbor;type II,arbor+shrub;type III,arbor+sub-arbor and type Ⅳ,arbor+sub-arbor+shrub.Their width varied among 4~10 m.The noise-reducing ability (y,dB) of the green belts varied from 0.93 to 12.96 dB with various belt widths (x,m),and can be expressed as following:y=1.2251x+0.2416.Meanwhile the additive attenuation of traffic noise (y,dB) presents a closely linear relationship with the total noise-reducing ability (x,dB) as following:y=0.4535x+0.2698 (R2=0.9242).Meanwhile,the additive attenuation of traffic noise is main affected by the constructions of green belts,which reached 0.93,2.25,4.43 and 6.72 dB,respectively for the above four kinds of construction types.It shows that the width and construction of green belts play the same important role to reduce the traffic noise.
