    Making Template DNA in the Analysis of Tobacco Germplasm Based on AFLP Fingerprint

    Two methods-CTAB method and SDS method were tested in isolation of tobacco genomic DNA. The results showed that the degree of purity of DNA extracted by using CTAB method was higher than that by using SDS method; the value of OD260/OD280 for the DNA samples obtained with CTAB method was over 1.8; the molecular weight of DNAs was more than 23kb, the DNAs were completely digestible with EcoRⅠ/MseⅠ, and the AFLP figure were obtained.

    Cytohistological Studies on Morphogenesis Adventitious Buds in Tissue Culture of Lilium longiflorum

    Sterilized leaf segments of lilium longiflorum were cultured for callus inducement and the results showed that MS+6-BA 2.0mg/L+NAA 0.2mg/L was the best for adventitious buds. The leaf segments were dissected at regular intervals to observe the initial places of dedifferentiation and histological changes during induced period. The results shows that the dedifferentiation initiates from wounded cells; the dedifferentiated cells form meristematic packed cells which were developed to be callus; the callus dedifferentiates and the adventitious buds form. The buds mainly develop from second to seventh layer cells of meristematic packed cells.

    The effect of manganese (Mn) on root activity, contents of proline, protein and malonaldehyde in leaves were studied using method of water culture. The results showed that appropriate Mn increased root activities of soybean and decreased contents of proline, protein and malonaldehyde in leaves, but the result was reversed under high Mn. There was difference in the response of soybean to Mn between Zhechun No.2 soybean and Zhechun No.3 soybean, and Zhechun No.3 was more sensitive than Zhechun No.2.

    Comprehensive Identification and Vigor Comparison of Agrobacterium rhizogenes

    The comparison and identification of Agrobacterium strains (A4,R1205, R1000, R1601, R1022 and 15834) were investigated by 3-ketolactose production, differential acid production and motility test. The result indicated that R1205, R1601, R1000and A4 belonged to biotype Ⅱ and the vigor of strains was R1000 > R1205 > A4 > R1601. According to PCR assay, four strains (A4, R1205, R1000, R1601) were Agrobacterium rhizogenes. According to the transformatic model cucumber, which mediated by A4, R1205, R1000 and R1601, the transformatic frequency of R1000 was the highest (79.6%), and followed by R1205, A4 and R1601. Based on the experiment results, the vigor of R1000 was the highest.

    Effects of Chemical Treatment and Bagging on Fruit Quality of Cara Cara

    The experiment was performed to study the effect of NaCl, MnSO4, NaH2PO4, GA3 treatment and bagging fruits after the second physiological fruit abscission on quality of Cara Cara fruit. The results showed that GA3 treatment increased the index of fruit shape, bagging could improve the rind brightness and yellowness extremely remarkably, but bagging without bag removal before fruit coloring decreased rind redness and weight of single fruit extremely remarkably; NaH2PO4 treatment increased the total of soluble solid remarkably, NaH2PO4 and GA3 treatment improved TA and decreased the ratio of TSS/TA. Bagging and bag removal before fruit coloring and MnSO4 treatment increased the total of soluble sugar extremely remarkably, bagging and bag removal before fruit coloring and NaH2PO4 treatment increased VitC content remarkably.

    Effects of Water Stress on Pollen Viability and Storability of Macadamia

    Under water stress treatments, the viability and storability of pollens collected from 3 varieties (Kau, Pahala and O.C.) of Macadamia were studied. The results showed that the viability of fresh pollen decreased with the storage time, and at room temperature the pollen could be stored for 6 days. Pollen viability decreased quickly after storing 6 days under water stress, with the increase of water stress, the pollen viability decreased more quickly. Under the different water stress treatments (i.e. saturated irrigation treatment, normal irrigation treatment, deficient irrigation treatment and CK), the storage life of pollen was 6d, 30d, 10d and 4d, respectively. The storability of 3 varieties was Kau >Pahala >O.C. and the best way to identifying pollen viability was biphenyl amine phenol.

    Cultured Narcissus tazetta var. Chinenese in the solution added with mineral element, it was found that mineral nutrition could significantly improve the net photosynthetic rate and the ratio of photosynthesis to respiration, increase the content of soluble protein and chlorophyll in leaves in the stage of sprout and flowering period, as well as reduce the activities of SOD, POD and CAT, the production rate of O2· and MDA content of leaves in the stage of wilting. Mineral nutrition could effective prevent the over-oxidazition of membrane and the senescence of leaves.

    Application of the Principle Component Analysis on Mango Storage Characteristic

    The principle component analysis(PCA) was used for statistic analysis on eight direction signs of post-harvest mangoes. The results showed that all the direction signs were vested in tow parts:one was storage characteristic factor, the other was physiological factor. The storage characteristic was composed of VitC content, titratable acid content, losing rate and chlorophyll content, and the physiological factor was composed of SOD activity, LOX activity and MDA content. Their dedicative rate was 63.69% and 32.46%, respectively, and the accumulative proportion for the tow factors was 96.16%. It reflected the information for all direction signs essentially. The integrated evaluate model on these basics also showed that the storage quality was improved a bit within 10d and then declined rapidly. These reflections were coincident with the facts basically.

    Effects of Municipal Domestic Sewage Effluents on the Growth of Calendula officinalis

    The seeds and potted plants of Calendula officinalis were treated with municipal domestic sewage effluents. The treatments were divided into 4 groups according to effluents:tap water(v/v):I, 1:0; Ⅱ, 1:0.5; Ⅲ, 1:1; control, 0:1. Compared with the control, sewage irrigation treatments (I, Ⅱ and Ⅲ) obviously reduced the rate of seed germination and increased the disease incidence of seedlings. The undiluted raw sewage could remarkably restrain the seedling growth, and the retarding effect could be relaxed by diluting with tap water. Furthermore, the diluted sewage of proper concentration could improve the seedlings growth. Compared to the control, sewage irrigation treatments increased the fresh weight of the stem, leave and root of full-grown C. officinalis plant by 161.63%~215.12%, 86.77%~109.23% and 23.89%~34.13%, respectively. The results of this study showed that the diluted sewage could be reused in irrigation and the nutrient elements in the sewage effluents could increase the ornamental value of herbage flowers.

    Effects of Weeding Compound Fertilizer “Dao-shui-bao” on Rice Production

    Field experiments of fertilizer application treatments (Ⅰ, weeding compound fertilizer ‘Dao-shui-bao’;Ⅱ, mixed fertilizer of 50% Butachlort+formulated fertilizer; Ⅲ, mixed fertilizer of 10% Bensulforon methyl+formulated fertilizer; Ⅳ, mixed fertilizer of 50% Butachlor+10% Bensulforonmethyl+formulated fertilizer; Ⅴ, CK) on rice were conducted. All the fertilizer application had the same concentrations of N, P2O5 and K2O. The results showed that treatment Ⅰ significantly increased the weeding efficiency and rice yield compared to the treatment Ⅲ and treatment Ⅳ, there were no significant differences between the treatment Ⅰ and treatment Ⅱ in rice production. The results also suggested that application of weeding compound fertilizer ‘Dao-shui-bao’ will result in savings of energy, reducing the costs and increasing profits, and is safe in rice production.

    Karyotype Analysis of Two Buckwheat Variety Chromosomes

    The karyotype of two varieties of buckwheat GaoQiao 3 and XiQiao 5 were studied. The results showed that both the two varieties of buckwheat belong to ‘2A’ type. The karyotype of the buckwheat GaoQiao 3 is 2n=2x=16=10m(2SAT)+4sm+2st and the karyotype of the buckwheat XiQiao 5 is 2n=2x=16=8m+6sm(2SAT)+2st.

    A TLCS method for determining the content of ursolic acid in Clerodendranthus spicatus was established in this experiment. The method was as follows. After extraction, the samples were dripped on silica gel GF254 TLC plate, using a mixture of the alkane-chloroform-acetic acid B ester-methyl alcohol (25:15:5:2) as developer and locating the position under the light of an UV lamp(254nm); double wavelength reflection sawtoothed scanning was performed λS=520nm, λR=650nm. The results showed that the regression equation is y=3658.34 X-7857.07, r=0.9998 and linear range is 0.5~2.5μg. The average recovery rate is 101.57%, RSD=2.83%.

    Extraction of Dicranopteris dichotoma Polysaccharides and Its Antibiotic Activities

    Dicranopteris dichotoma polysaccharide was extracted by four methods,among those that the extracting rate of synthesis method was the highest,reaching 5.2%. The crude polysaccharide was purified by DEAE-52 and the neutral polysaccharide was eluted by distilled water and the polyanion polysaccharide was gradually eluted by 0.8mol/L NaCl. Their contents were 78% and 22%, respectively. The antibiotic activities of Dicranopteris dichotoma were tested with the agar disc diffusion method. The result showed that the polysaccharide had various amount of antibiotic activities against bacteria and fungi.

    Taking a sample of six early-maturing, tasty Eurasia grape varieties for experiment, their phenological phase, fruit economic properties, disease resistance, secondary fruit growth and yield under the equipped circumstances of rain prevention were studied. The results showed that among these six varieties there were obvious differences in their yield, quality and disease resistance. By comparison of comprehensive properties, Jing-Xiu is the best and the following one is Ogoust, and these two varieties are valuable for popularization in Lingde City.

    Isopentenyl-transferases catalyze the first and rate-limiting steps of cytokinin biosynthesis, and the corresponding genes have been cloned. A family of genes from Arabidopsis coding for cytokinin biosynthesis enzymes have been identified by a bioinformatic approach. It is speculated that these genes might be expressed in distinct spatial and temporal patterns to regulate cytokinin biosynthesis. This review specially introduced the functions and advances of ipt in cytokinin biosynthesis.
