    Root Development in Arabidopsis Folylpolyglutamate Synthetase Mutant adtfb under Low-nitrogen Condition

    T-DNA insertion mutant of folylpolyglutamate synthetase atdfb was used to explore the role of folates during root development in Arabidopsis. Phenotype of three-week-old atdfb seedlings was similar to the wild type, however, the mutant displayed shorter primary root than the wild type on 1/2MS, and phenotype of complemented seedling was recovered indicating that the shorten root phenotype was due to the loss function of AtDFB. Length of primary root of 11-day-old atdfb-3 seedlings was 23% of the wild type on 1/2MS medium that was nitrogen (N)-sufficient conditions. The development of atdfb-3 almost stagnated under low-N conditions, and the primary root length of 11-day-old atdfb-3 seedlings was only 4% of the wild type. Under low-N conditions, exogenous 5-formyl-tetrahydrofolate (5-F-THF) restored the phenotype of atdfb-3 to the wild type. With the application of 5-F-THF, the primary root length, root hair length and the quiescent center cell organization in atdfb-3 were recovered. This study indicated the necessity of folates in Arabidopsis root development and adapting to low-N condition.

    An Efficient Plant DNA Extraction and PCR Amplification System

    A simple and efficient system of plant DNA extraction and PCR amplification was established. The system only need one step to extract DNA, and good amplification results of target genes could be obtained by prolongation PCR predenaturation time. The system had strong experimental stability and universal species applicability. It would greatly improve the efficiency of plant molecular biology experiment and plant breeding experiment based on molecular marker, and decrease scientific research cost.

    Effects of Aluminum Toxicity and Acidity on Moisture Content of Gingko under Soilless Culture

    ‘U207’ even design was used to study the effects of acidity and aluminum (Al) on moisture content of Gingko under soilless culture. Results showed that moisture content of Gingko was decreased with the acidity increasing as well as the Al concentration, which drew a conclusion that Gingko root could not absorb water regularly under high acidity and Al concentration culture. The result also showed that pH was the most important factor impacting on Gingko growth, the plant could not grow when pH was below 3.5, but the plant growth could not impact by Al concentration when pH was higher than 5.0. Therefore, Gingko plant moisture content could be as evaluation index under acidity and Al stresses.

    Physiological Characteristic and Hardness Changes of Agaricus bisporus under Low Temperature Preservation

    The changes of physiological quality of Agaricus bisporus after storage at 4 ℃ for 3 d, 6 d, 10 d and 14 d were studied. The results showed that the hardness of A. bisporus decreased slowly under 4 ℃ preservation. Degradation of soluble protein and POD, SOD, CAT activity were slower. The content of V? decreased rapidly on the 3rd day and then maintained stable level. The level of MDA, which had remained stable, rose rapidly on the 10th day. The results showed that low temperature preservation had positive effect on quality of A. bisporus during storage and could prolong its shelf life.

    Effect of pre-cooling and storage temperature on fresh bamboo shoot aging of Bambusa oldhamii was studied. Results showed that aging speed, preservation period and weight loss ratio of B. oldhamii shoots were affected obviously by pre-cooling and storage temperature. Pre-cooling treatment could restrain aging of fresh bamboo shoot, reduce aging rate and prolong preservation period. The aging rate and weight loss ratio of fresh bamboo shoot under (5±0.5) ℃ constant temperature chamber was lower than those of (30±0.5) ℃ constant temperature chamber, but its storage period was on the contrary. When fresh bamboo shoots stored 3, 6, 9 and 12 days, the weight loss ratio of fresh bamboo shoots at (5±0.5) ℃ constant temperature chamber was lower than 1%, which was 26.32%, 38.10%, 48.04% and 55.23% respectively at (30±0.5) ℃ constant temperature chamber.

    Population Structure and Dynamics of Endangered Plant Tetracentron sinense

    Based on population life table and theory of survival analysis, the individuals of Tetracentron sinense in Caopo Nature Reserve of Sichuan Province were graded according to the size of diameter at breast height, and the age structure was represented by tree diameter structure. The static life table of T. sinense population was compiled by smoothing out technique. The mortality rate curve, killing power curve, survival rate curve, and survival function curve were also analyzed to reveal the quantitative characteristics of T. sinense population. Additionally, quantitative dynamics of the population was predicted using Time Series Model. The results showed that: (1) The age structure of T. sinense in Caopo NR was stable and the seedlings were abundant, but the mortality of sapling was relative higher, indicating the recessive characteristic of the population. (2) The mortality rate curve and killing power curve of T. sinense population in lifespan had similar variation trend, which had a peak in third age-class, i.e. the sapling mortality rate was higher and the survival curve of population tended to Deevey-Ⅱ. (3) The four survival function curves showed that T. sinense population decreased quickly in early age period, and became stably in middle age period, but declined in old age period. (4) Time Series Forecasting Model showed that population quantity of T. sinense decreased in the next 9 age-classes, the number of aged individuals gradually increased, the young plants was decreasing, indicating the recessive character of the population.

    Population Structure and Spatial Distribution Pattern of Endangered Species Rosa praelucens

    In order to reveal population characteristics of Rosa praelucens, 10 sample plots were set in its distribution area in Shangri-La, and life table, survival curve, χ2, AC, PC of R. praelucens were compiled to analyze the population structure of R. praelucens. CA, I, K, PAI and other aggregation intensity indexes were used to analyze the population distribution pattern so as to provide theoretical basis for population protection of R. praelucens in Shangri-La. Results showed that: (1)R. praelucens belonged to the recession, with few young trees, large proportion of elderly individuals, and slow population renewal. (2)According to Deevey classification, the survival curve of R. praelucens belonged to Deevey III. (3) Through χ2 test, it was found that the independence of individual species in R. praelucens community was relatively large. (4) The distribution pattern of R. praelucens population basically followed the pattern of aggregation distribution.

    There were 245 species belonging to 199 genera in 99 families from the limestone area of Yinde. The numeral dominant families were Euphorbiaceae, Papilionaceae, Orchidaceae, Pteridaceae, etc. The geographical elements of family and genus were tropical and sub-tropical. There were 7 endemic genera to China, such as Eurycorymbus and Pteroceltis, 15 endemic species to limestone area, such as Sapium rotundifolium and Zenia insignis, 15 species belonging to 5 families and 13 genera of rare and endangered species, such as Eurycorymbus cavaleriei. The dominant species of vegetation were Dimocarpus longan, Sterculia lanceolata, Adenanthera pavonina var. microsperma, and Cinnamomum burmanni, etc. Based on the special habitat of limestone, the protection on plant diversity should be strengthened.

    Adaptability Assessment of 40 Varieties of Crassulaceae Succulents in Zhuhai Area, Guangdong

    With the aim of selecting varietal resource of Crassulaceae Scculents for landscaping in Zhuhai area, 40 varieties of Crassulaceae Scculents had been planted in opened field for two years. Observations of the growth were made, and ornamental characteristics as well as ecological adaptability were evaluated. Results showed that 9 varieties were categorized as Ⅴ grade, including Graptopetalum ‘Bronze’, Graptopetalum paraguayense, Sedum adolphli, Graptosedum ‘Francesco Baldi’, Kalanchoe marmorata, Kalanchoe ‘Fortyniner’, Crassula portulacea, and Graptopetalum mendozae and Graptopetalum ‘Mirinae’. 6 varieties were categorized as Ⅳ grade, including Orostachys furuser, Echeveria runyonii, Echeveria huthspinke, Pachyphytum oviferum ‘Pachytoides’, Pachyphytum ‘Doctor Comelius’, Sedum super ‘Burros’. Above varieties were suitable for cultivation, popularization and application in Zhuhai area.

    Full investigation including plant application type, ex-situ conservation and science popularization education in shade-tolerate botanical area of Shenzhen Fairylake Botanical Garden was conducted. The results showed that there were totally 520 species belonging to 268 genera of 132 families applied in shade-tolerate botanical area, of which 20.0% trees, 11.7% shrubs, 60.2% herbs, and 8.1% vines; 387 protophytes were ex-situ conservation, accounting for 74.4% of total plant species. Science popularization education was mainly displayed by billboards and nameplates.

    PSO-LSSVR-Based Soil Fertility Evaluation

    Associated with Particle Swarm Optimization (PSO) and Least Squares Support Vector Machine (LSSVR), this study proposes a soil fertility evaluation model based on PSO-LSSVR. Ten soil indicators were selected as elevation indexes, which included organic matters, total nitrogen, available phosphorus, available potassium, cation exchange, capacity, acidity-alkalinity, volume weight, clay dispersion, water stable aggregated and dispersion ratio. The black soil in Jilin province was used example to construct soil fertility evaluation model. The result was compared with two other methods (extension-element method and ordinary SVM method), the soil fertility levels of most samples were consistent, sample 2 and sample 13 were classified as grade Ⅳ and grade Ⅲ by PSO-LSSVR model respectively, which were in line with the actual situation, which showed that PSO-LSSVR model was a suitable soil fertility evaluation model and the result accurately reflected soil properties.

    Newly discovered vascular plants in Fujian province reported in literatures from 2015 to 2017 were statistically analyzed, including 11 new species (5 families) and 80 new record species (35 families). So there were 5064 species of vascular plants in Fujian, belonging to 1643 genera in 252 families.

Chen and Anoectochilus zhejiangensis Z. Wei et Y. B. Chang, all of which were discovered in the southern slope of Mufu Range region. And the floristic geographic implications of those new record species were discussed.

    Effects of different disinfection treatments, additives, plant growth regulators on tissue culture and rapid propagation system of Habenaria rhodocheila with seeds as explants were studied. Sterilizing the seeds with 0.1% mercuric chloride for 15 min was the best disinfection treatment. Seeds germinated only on the medium adding active carbon(AC). 0.2% active carbon in culture medium was fit for germination. The optimal medium for seed germination was 1/2MS+NAA 0.5 mg·L-1+AC 0.2%, the optimal medium for multiple buds formation was 1/2MS+NAA 0.2 mg·L-1+ZT 3.0 mg·L-1+AC 0.2%, the optimal rooting medium was 1/2MS+NAA 0.1 mg·L-1+6-BA 2.0 mg·L-1+AC 0.2%.

    Tissue Culture Technology of Cymbidium hybridum

    To set up a new tissue culture system with the buds of Cymbidium hybridum as tissue explants, N6, B5, CC and MS as basic medium, orthogonal experiment design was carried out with macroelement, microelement and organic matter as three different influencing factors. Results showed that the best medium combination for developing C. hybridum was B5 macroelement + CC microelement + MS organic + sucrose 30 g·L-1 + agar 7 g·L-1 + activated carbon 0.5 g·L-1 (pH 5.8—6.0), the best medium hormone concentration combination for multiple bud proliferation was 6-BA 4.0 mg·L-1+ NAA 0.2 mg·L-1,the best hormone concentration for induce in vitro shoot rooting was NAA 1.0 mg·L-1.

    Comparison of Cutting Yield of Pinus taeda under Different Managements

    In order to build a high-yield cutting nursery of Pinus taeda, the 3-level L9 (34) orthogonal test was conducted. The results showed that the optimum combination of cutting yield in spring was 15 cm truncation height, row spacing of 50 cm × 80 cm; in summer, the optimized combination was farmyard manure + compound fertilizer, 15 cm truncation height, row spacing of 50 cm × 80 cm; in autumn, the optimized combination was farmyard manure + compound fertilizer, row spacing of 50 cm × 80 cm, and 12 cm truncation height; in winter, the optimum combination was farmyard manure, 15 cm truncation height, and row spacing of 40 cm × 50 cm. Improved combination of annual cutting yield was farmyard manure + compound fertilizer, 15 cm truncation height, and row spacing of 40 cm × 50 cm. Therefore, there should be used different management measures in different periods for increasing cutting yield of Pinus taeda.

    Effect of Supplemental Lighting on Promoting Flower of Pitaya in Greenhouse in Winter

    Effect of supplemental lighting on promoting flower of pitaya in greenhouse in winter was carried out. Results showed that the flower bud rate of pitaya reached upto 83.3% in the first batches at the conditions of average light intensity 500 lx, 6 h each day for lasting 45 d, the average flower bud rate of old branches(branch age was equal or greater than twelve months) was 15.2%, the average flower bud rate of young branches (branch age was less than twelve months) was 28.0%, the flower bud branches occupied 21.4% of all branches. The percentage of bud points which flowered on old branches was 2.7 percent, while that on young branches was 2.9%. However, there was not flower bud on those plants without supplemental lighting. The results indicated that supplemental lighting could effectively promote flower buds of pitaya in greenhouse in winter.

    Citrus depressa was introduced to Xiamen area. The adaptive cultivation experiment was performed to evaluate the biological characteristics, growth trait, fruiting and nutritional ingredient as well as ecological habits of C. depressa. Results indicated that C. depressa had good adaptive capacity in introduced area. The plant fast grew, flushes of reproductive shoots occurred 4—5 times each year with several batches of flowering and fruiting, the yield of fruits was up to 5460 kg·hm-2 and the nobiletin and tangeretin contents were high in the fruit.

Recently quinoa is worldwide popular for its totally nutritious qualities, stress-tolerant characteristics, which lead quinoa from an Andean food to a strategic resource. Nutrition, adaptation, second metabolites, medicinal properties, germplasm preservation, utilization and development strategies of quinoa production were reviewed.
