    Community Structure of Phytoplankton in Nanji Islands Sea Region in Spring and Summer 2006

    The species diversity and cells density of phytoplankton and their variations in Nanji Islands Sea Region were investigated in 2006(from April to June).Totally,68 taxa of 4 phyla were observed,including 53 taxa in Bacillariaphyta,13 taxa in Dinophyta,1 taxon in Cyanophyta and 1 taxon in Chrysophyta.Along with different investigation periods,the main dominant species of phytoplankton changed.The main features of phytoplankton are characterized by the occurrences of Prorocentrum triestinum bloom and P. donghaiense bloom in middle May and later May,respectively.The formation of Prorocentrum bloom has close relation with water temperature and causes the corresponding changes of nutrient structure of seawater.

    Total DNA Extraction and RAPD System Optimization of Fortunella crassifolia

    Three different methods were used for total DNA extraction from fresh leaves of Fortunella crassifolia,respectively.The results showed that the most effective method for RAPD analysis was improved CTAB Ⅱ protocol.Four influencing factors on RAPD-PCR,including Mg2+,dNTP,primer and DNA template concentration were tested using single factor design and uniform design.A suitable RAPD-PCR system for F. crassifolia was established as follows:20 μl PCR solution,2.5 mmol/L Mg2+,0.25 mmol/L dNTP,0.20 μmol/L primer,1.0 ng/μl DNA template,1 U Taq DNA polymerase and 10×buffer.The RAPD-PCR was programmed by a cycle for 3 min at 94℃,followed by 45 cycles for 45 s at 94℃,60 s at 37℃ and 120 s at 72℃,and finally by a cycle for 10 min at 72℃,storing at 4℃.

    Effects of Nitrogen Application and Cotyledon Excision on Leaf Nitrate Reductase Activity of Swietenia macrophylla Seedling

    Under nitrogen application and cotyledon excision,the nitrate reductase activity (NRA) of seedling leaf of Swietenia macrophylla,a tropical defoliated arbor of Meliaceae,was studied.The results showed that the leaf NRA increased first but decreased then with the development of leaves under non-nitrogen application.After nitrogen application,the leaf NRA of seedlings of all sampling periods (except 35 days after transplanting) were significantly higher (P<0.05) than that of the control and a gradual decrease was observed during the development period of leaves.The leaf NRA of S. macrophylla seedling increased significantly (P<0.05) after 4 weeks of cotyledon excision during different development periods.

    Physiological Effects of 6-BA and GA3 on Flower Bud Differentiation of Nobile Type Dendrobium

    Based on the investigation of pseudobulbs horizontal elongation and growth of nobile type dendrobium,the effects of growth regulators such as GA3,6-BA on the floral bud differentiation were studied.The results showed that the rapidly thickening stage of the pseudobulbs were from September 15th to October 10th,about 50 d.The growth regulators of 6-BA,6-BA+GA3 improved the floral bud differentiation of nobile type dendrobium during this period.Three different concentration treatments of 6-BA promoted floral bud differentiation significantly,and the concentration of 400 mg/L was the best.Proper concentration of 6-BA plus GA3 also promoted flower bud initiation.The optimum concentration of the three treatments was 6-BA 200 mg/L+GA3 50 mg/L.The effects of GA3 on the flower bud initiation were not steady.The defoliation phenomena was not obvious under GA3 concentration below 25 mg/L while large numbers of leaves fallen off with GA3 concentration above 50 mg/L.

    Effect of Salicylic Acid on Banana Seedlings under Chilling Stress

    Bananas seedling leaves were sprayed by 0.6 mmol/L salicylic acid (SA),then in cold storage at 8℃ for 5 d.The effects of SA on resistance of banana seedling to chilling injury were studied.The results showed that pretreatment with 0.6mmol/L SA reduced the contents of MDA and hydrogen peroxide,and the relative ratio of electrolyte,retarded the decreasing of soluble protein,increased the content of proline and the activities of POD and CAT in banana seedling leaves.It indicated that SA could increase the cold tolerance of banana seedling,which was mainly achieved by means of enhancing the anti-oxidative enzyme activities,inhibiting the accumulation of reactive oxygen and decreasing the injury of membranes under cold stress.

    Effects of Selenium on Growth and Anti-oxidative Enzyme Activities in Pueraria phaseoloides Hairy Roots

    Effects of selenium (Se) on the growth and anti-oxidative enzyme activities of Pueraria phaseoloides hairy roots were investigated.The results showed that low concentration of Se (0~0.5 mg/L) promoted the growth and isoflavones biosynthesis of hairy roots,and high concentration of Se inhibited the growth and isoflavones accumulation,and the inhibition effects increased with the concentration of Se.In addition,Se also promoted the biosynthesis of soluble protein,and the effects increased with the concentration of.Meanwhile,Se decreased the activity of SOD and content of MDA,enhanced the activity of POD.It was suggested that Se play an important regulative role in the growth and antioxidant enzyme activities of P. phaseoloides hairy roots.

    Eight agricultural characteristics of 15 germplasm accessions of legume pasture that introduced from Australia were evaluated comprehensively by using gray relation analysis,membership function analysis,cluster analysis and principal component analysis.The applications of the four statistical methods on comprehensive evaluation for legume germplasms were discussed.The evaluation results showed that stylo Nina,stylo Temprano,stylo 540 and stylo 541 were the better varieties,and the results of four statistical methods for Legume germplasm were in substantial agreement,so that the four statistical methods can be used for comprehensive evaluation of legume germplasm independently.

    Aseptic test-tube plantlets of Alocasia amazonica were gained from shoot tip culture.The effects of hormone and coconut milk combinations on proliferation of Alocasia amazonica and the effects of hormone and MET combinations on rooting were studied.The suitable transplanting substrate for test-tube plantlets of Alocasia amazonica was screened.The results showed that the suitable culture medium for proliferation of Alocasia amazonica was MS+6-BA 4.0 mg/L+NAA 0.2 mg/L+CM 15%~20%.The suitable culture medium for rooting of Alocasia amazonica was 1/2MS+NAA 0.5 mg/L+MET 0.5 mg/L.The best transplanting substrate was perlite and the survival rate was up to 100%.

    By the methods of gas chromatography and the field demonstration experiments,and according to the Maximum Residue Limit (MRL) of dichlorvos,profenofos and butachlor of GB2763-2005,Japan and European Union,the securities of dichlorvos,profenofos and butachlor in vegetable-soybean were studied.The results showed that the pre-harvest interval of dichlorvos was 3d,and the product was safe when dichlorvos was rationally applied in vegetable soybean.The pre-harvest interval of profenofos was 18d (reference cabbage) in China and was 28 d in Japan and European Union.When profenofos was improperly applied in the late middle growth stage or late growth stage of vegetable-soybean,the pesticide residue in final product was beyond the standard of maximum residue limit in food.With proper application of butachlor at the day of vegetable-soybean seeds being sowed,the final product was of safe standard.

    Based on field investigation,the species compositions of plant community in Xiaoyanwan garden of Xingwen block-sea were analysed.There are 90 species plants belonging to 82 genera of 48 families,among which the species of Tiankeng is the richest,75 species;the three corroded depressions have 52 species;and the species in Monkey Lake is the least,only 12 species.Shannon diversity index was used to do a comparative study on the species diversity of the five plant communities.The Shannon index H' of the five plant communities is ranked from high to low as follows:4.056 of Tiangkeng,2.882 of the No.1 corroded depression,2.878 of the No.2 corroded depression,2.821 of Monkey Lake,and 2.258 of the No.3 corroded depression.Some suggestions for further researches on the vegetation and plant community in deserted regions and some protection efforts for the vegetation in this area were put forward.

    To provide a scientific basis for reforestation,the important value,Shannon-wiener index,Simpson index,richness index,Pielou evenness index and Sorensen similarity coefficient were applied to study the plant diversity in planning afforestation of Yuntai Mountain in Lianyungang.The results were as follows:Phyllostachys varioauriculata was the most in shrub layer,and the important value of Carex pallens was the highest in herb layer.Most plant diversity indexes of herb layer were higher than those of shrub layer basically.Sorensen similarity coefficients showed that the nearer the research areas,the more the same plant species they shared.Shrub plant diversity was related to soil thickness and exposure.The elevation and exposure had some effects on plant diversity in herb layer.

    The floristic characteristics of Machilus versicolora community in Mangdangshan Nature Reserve in northern Fujian were studied with the methods of typical investigation and routine survey.The species diversity is rich in this community and the regional families and genera dominate with single species and less species.The dominant families are Lauraceae,Theaceae,Rubiaceae,Rosaceae,Myrsinaceae and Moraceae.The Machilus, Camellia, Ilex and Ficus are the dominant genera and the typical families are Lauraceae,Theaceae,Moraceae,Myrsinaceae,Araliaceae and Aquifoliaceae.The flora is ancient and scattered with obvious dominant families and genera,and shows the feature of south China flora.

    Investigation was carried out on vegetation resources in Jiedong Sangpu Shuanglu Nature Reserve by route survey and typical sample plots.There are 580 species of vascular plants,belonging to 129 families,371 genera.The tropical and sub-tropical taxa are predominant,taking up 75% of seed plant flora.The original vegetation is sub-tropical evergreen broad-leaved forest,composing by communities of Lithocarpus and Cyclobalanopsis,but its percentage of area is not great.The area of Pinus massoniana community and Acacia confuse community is large.It is concluded that the activities of human have great impact on the composition and structure of forests.Hill closing for reforestation and stand improvement are recommended,helping the succession from coniferous forests and coniferous and broad-leaved forests to region-specific climax communities.

    Botanical gardens exist and development on the basis of plant resources,and therefore conservation of plant resources is primary work of botanical gardens.Previous conservation of plant resources mostly included maintaining and reproduction of plant resources,and researches on conservation of plant resources.This model will face more and more limitations when botanical gardens either develop to a certain extent or meet more and more challenges.Innovative ideas on conservation of plant resources are to systematically choose plant resources to conservation and to utilize plant resources conserved by botanical gardens.Utilization of plant resources includes fundamental researches and application researches on plant resources,popularization and exploitation of plant resources,and popularization of science by plant resources.
